dc.description.abstractThe marine environment is extremely vast and encompasses a multiformity of different habitats, hosting most of the planet's biodiversity. Some of these, although particularly significant from an ecological point of view, are practically unknown from a microbiological point of view due to their extreme conditions. In this work, the bacterial communities of some extreme or sub-extreme marine environments, practically unknown from a microbiological point of view, have been characterised at taxonomic and structural levels: an arctic intertidal environment, Kandalaksha Bay (KB), Russia; a hypersaline transition area, Saline di Tarquinia (ST), Italy; an ocean trench, Kuril Kamchatka Trench (KKT), Pacific Ocean. The metagenetic analysis of the bacterial communities of water samples selected in relation to the different parameters characterising the environments (depth, salinity and tidal currents) was performed. The amplicon libraries for the NGS sequencing (pyrosequencing 454 and Illumina MiSeq) were prepared from the DNA obtained from the water samples. The structure and the composition of the bacterial communities were analysed. The KB community, an area characterised by strong tidal currents, were rather similar, especially in the surface layers. In fact, the analyses carried out showed that the only great differences were found in the deepest sample, not influenced by the tidal currents. In addition, the main parameters structuring the communities were water temperature and salinity. As for the ST salterns, along the salinity gradient the communities developed in a rather clear way, if compared with the adjacent sea waters (feeding the ponds). The community variations in addition to the salinity were attributable to the sampling site and period. Unique communities have differentiated in the brine storage basin (BSB), having remained isolated from the the adjacent ponds for decades. The salinity was the only parameter structuring the BSB communities. Overall, some taxa were found into a large range of salinity. The investigation carried out on the KKT samples showed that the bathyal and abysso-hadal communities were structured according to the depth. A high number of unassigned OTUs were obtained for the abysso-hadal samples; even considering the limits of the methods used, it suggested that the bacterial diversity of this environment is still poorly understood. Moreover, it was confirmed that the composition of the deep-sea bacterial communities can vary between geographic areas distant from each other but also within the same study area.it
dc.description.abstractL’ambiente marino è estremamente vasto e racchiude una multiformità di habitat differenti, ospitando la maggior parte della biodiversità del Pianeta. Alcuni di questi, sebbene particolarmente significativi a livello ecologico, sono praticamente sconosciuti dal punto di vista microbiologico, date le loro condizioni estreme. In questo lavoro sono state caratterizzate, a livello tassonomico e strutturale, le comunità batteriche di alcuni ambienti marini estremi o sub-estremi, che dal punto di vista microbiologico risultano praticamente sconosciuti: un ambiente artico intertidale, Kandalaksha Bay (KB), Russia; un’area di transizione iperalina, Saline di Tarquinia (ST), Italia; una fossa oceanica, Kuril Kamchatka Trench (KKT), Oceano Pacifico. È stata condotta l’analisi metagenetica delle comunità batteriche di campioni di acqua selezionati in relazione ai diversi parametri caratterizzanti gli ambienti (profondità, salinità e correnti di marea). A partire dal DNA ottenuto dai campioni d’acqua sono state preparate le librerie di ampliconi per il sequenziamento NGS (pirosequenziamento 454 e Illumina MiSeq). Considerando i dati di sequenza, sono state analizzate struttura e composizione delle comunità batteriche. La comunità di KB, area caratterizzata da forti correnti di marea, risultavano piuttosto simili soprattutto negli strati superficiali. Infatti, le analisi effettuate dimostravano che le uniche grandi differenze si riscontravano nel campione più profondo, non influenzato dalle correnti di marea. Inoltre, i parametri principali strutturanti le comunità erano temperatura dell’acqua e salinità. Per quanto attiene le ST, si riscontrano comunità che si sviluppano lungo il gradiente di salinità in maniera piuttosto netta, se comparate con le adiacenti acque del mare che alimentano le vasche. Inoltre, le variazioni di comunità oltre alla salinità sono imputabili al sito e al periodo di campionamento. Comunità particolari si sono differenziate nella vasca di stoccaggio delle salamoie isolata da decenni dalle vasche attigue. La salinità è risultata essere l’unico parametro strutturante le comunità. Nel complesso, si sono riscontrati evidenti fenomeni di adattamento di alcuni taxa alle ampie variazioni di salinità. L’indagine effettuata sui campioni prelevati lungo il KKT ha evidenziato che le comunità batiali ed abisso-adali si strutturano in funzione della profondità. Per i campioni abisso-adali del KKT si sono rilevati non comuni alto numero di OTU non determinate a livello tassonomico, che seppur considerando i limiti dei metodi utilizzati, fanno ipotizzare che la diversità batterica di questo ambiente sia ancora poco conosciuta. Inoltre, si conferma che la composizione delle comunità batteriche dei mari profondi possono variare, anche in modo consistente tra zone geografiche distanti tra loro, ma anche all’interno della stessa area di studio.it
