Abstract: 
Il carciofo (Cynara cardunculus L. var. scolymus L.) è una specie orticola perenne appartenente alla
famiglia delle Asteracee. La coltivazione è diffusa maggiormente nel bacino del Mediterraneo dove
ha avuto origine e in cui è presente la maggiore variabilità genetica. L’Italia è il primo produttore
mondiale di carciofo e il secondo esportatore dopo la Spagna. Tra le colture ortive da pieno campo il
carciofo occupa in Italia la terza posizione preceduto solo dal pomodoro e dalla patata.
Da un punto di vista nutrizionale, il carciofo ha un basso contenuto energetico e lipidico mentre è
ricco in proteine, sali minerali e fibra solubile e insolubile. Tra i costituenti del carciofo, le sostanze
fenoliche (acidi mono- e di- caffeilchinici e flavonoidi), gli zuccheri e l’inulina rivestono particolare
interesse sia dal punto di vista nutrizionale che tecnologico. Infatti, ai derivati dell’acido caffeico
sono attribuite proprietà epatoprotettive, coleretiche e diuretiche mentre ai flavonoidi attività
antinfiammatorie, antimicrobiche ed antiossidanti. L’inulina è un polimero del fruttosio che
promuove la crescita dei bifidobatteri della microflora intestinale (funzione prebiotica), contribuendo
a inibire la presenza di batteri dannosi. Ha inoltre effetti benefici per la salute, contribuendo a ridurre
il tasso di colesterolo e trigliceridi nel sangue.
Molti sono i fattori che possono influenzare il contenuto e il metabolismo di questi composti nel
carciofo e quindi la qualità del prodotto in termini di potenziale salutistico. Essi sono: genotipo,
organo considerato, stadio fisiologico, condizioni pedo-climatiche e agronomiche. L’interazione di
tali fattori rende quindi difficile l’approvvigionamento di materiale vegetale con caratteristiche
standardizzate.
Un approccio, dai costi accettabili, in grado di tenere sotto controllo i fattori non genetici è quello che
si avvale delle tecniche di colture in vitro. Nell’ambito di questo settore di ricerca si inserisce il
presente studio volto a sviluppare strategie biotecnologiche innovative per la produzione di molecole
bioattive (acidi caffeilchinici, flavoni ed antociani) e probiotiche (inulina) da colture cellulari e da
organi (radici, hairy roots) di un ecotipo tardivo di carciofo “Grato1”.
A tale scopo sono stati realizzati dei sistemi di coltivazione in vitro per la produzione di biomassa di
carciofo. In particolare, per la produzione ed analisi dei polifenoli sono state inizialmente definite le
condizioni sperimentali per l’allestimento di colture cellulari da tessuti fogliari di piantine in vitro e
successivamente gli estratti analizzati, con metodi spettrofotometrici e cromatografici (HPLC), per il
contenuto in polifenoli totali, in acidi caffeoilchinici totali e per l’attività antiossidante. I risultati
ottenuti, sono stati quindi confrontati con quelli presenti in bibliografia per il genotipo ‘Grato 1’
coltivato in pieno campo.
Per quanto concerne l’inulina, sono stati ottenuti e analizzati diversi tipi di materiale vegetale al fine
di individuare il sistema più idoneo alla sua produzione in vitro. In particolare, sono state allestite le
seguenti tipologie di colture: a) radici; b) callo da espianti radicali; c) “hairy roots” indotte mediante
infezione con Agrobacterium rhizogenens. Il contenuto di inulina dei materiali prodotti in vitro è stato
confrontato sia con quello di radici di piante di carciofo allevate in un sistema idroponico, realizzato
durante il dottorato, che con radici di piante allevate in campo.

The globe artichoke (Cynara cardunculus L. var. scolymus L.) is an ancient perennial plant species
belonging to the Asteraceae family, originating from the Mediterranean area, which today is widely
cultivated all over the word. Italy is the world's largest producer of artichokes and second largest
exporter after Spain. Among the vegetable crops, artichoke occupies the third position in Italy
preceded only by the tomato and potato.
From a nutritional point of view, artichoke has low energy and fat content and is rich in protein,
minerals and soluble and insoluble fiber. Among biochemical constituents of artichoke, polyphenols
(mono- and di- caffeoylquinic acids and flavonoids), sugars and inulin have an important role both
from the nutritional and technological point of view. In fact, it can be ascribed to the caffeic acid
derivatives hepatoprotective, choleretic, and diuretic properties while to flavonoid anti-inflammatory,
antimicrobial and antioxidant activity. The inulin is a fructose polymer that promotes the growth of
bifidobacteria in human intestinal microflora (prebiotic function), helping to inhibit the presence of
harmful bacteria. It also has beneficial effects on mineral absorption, blood lipid composition, and
prevention of colon cancer.
The levels of these compounds may vary according to globe artichoke genotype, organ considered,
physiological stage, pedoclimatic and agronomic conditions. The interaction of these factors does not
allow the supply of raw material with standardized features. The in vitro cultures have the potential
to control factors inducing variability in yields, making the process standardized and economically
sustainable.
The objective of this study was to investigate the possibility to produce bioactive (caffeoylquinic
acids, flavones, and anthocyanins) and prebiotic (inulin) molecules from cell cultures and organs
(roots, hairy roots) of artichoke “Grato1”. To this end, experimental conditions for the establishment
of in vitro systems for artichoke biomass production were defined.
In particular, for the production and analysis of the polyphenols, experimental conditions for the
establishment of artichoke cell cultures from leaf explants were defined. High-producing cell lines
were isolated and selected and analysed for total polyphenols and total caffeoylquinic acids content
and antioxidant activity. The results obtained were compared with those found in the literature for
genotype “Grato 1” grown in the open field.
Regarding inulin, to identify the most suitable system to its production in vitro, different types of
plant material were obtained and analysed. In particular, the following types of cultures were
established: a) roots; b) callus from root explants; c) "hairy roots" induced by infection with
Agrobacterium rhizogenens. The inulin content from in vitro materials was compared with those of
roots of artichoke plants grown in a hydroponic system and in the open field.
