dc.description.abstract | L’attività sperimentale condotta durante il triennio di dottorato è inserita in un progetto di ricerca a carattere interdisciplinare dal titolo “Studio della diffusione di inquinanti organici persistenti in ambiente rurale e stima dei rischi da esposizione per gli operatori del settore agricolo-zootecnico mediante modelli cellulari” finanziato dall’Istituto Nazionale per l’Assicurazione contro gli Infortuni sul Lavoro (INAIL). Obiettivo principale del progetto è analizzare i rischi da esposizione per gli operatori del settore agricolo-zootecnico che svolgono la loro attività nella Valle del fiume Sacco, un contesto agro-ambientale fortemente compromesso sotto il profilo della qualità e dello stato dell’ambiente, a seguito degli avvenimenti a carattere emergenziale connessi con la diffusione degli isomeri dell’esaclorocicloesano (HCH) potenti pesticidi appartenenti alla famiglia dei composti organoclorurati. Sulla base di quanto premesso, il lavoro oggetto della presente tesi è stato focalizzato su due obiettivi di ricerca. Inizialmente è stata valutata la qualità ambientale della Valle del fiume Sacco mediante opportuni campionamenti di varie matrici di rilevanza agro-ambientale effettuati nelle 9 aziende agrarie locali partecipanti al progetto. In particolare, è stata effettuata la verifica dei livelli di contaminazione da esaclorocicloesano (HCH) nel suolo e nel soprassuolo; nell’acqua per l’abbeveraggio degli animali in produzione; in alimenti d’origine animale destinati al consumo umano (latte, carne, miele, uova, ortaggi); in alimenti impiegati nell’allevamento dei bovini da latte. Pertanto, sono stati condotti una serie di campionamenti in varie zone all’interno dell’area di studio per garantire la rappresentatività delle varie matrici. Dopo una fase di pre-trattamento specifica per ogni matrice, ci si è avvalsi di metodi di estrazione accreditati a livello internazionale per l’estrazione degli HCH. Su tali campioni sono state eseguite le analisi gas-cromatografiche (GC-ECD). I risultati analitici ottenuti sono stati confrontati con i valori massimi consentiti dalla normativa vigente nazionale ed europea; inoltre, sono stati oggetto di una serie di elaborazioni statistiche per correlarli, per ciascuna azienda a diversi fattori (collocazione geografica, periodo di campionamento, profondità del suolo …). Tali risultati hanno evidenziato un livello di contaminazione ambientale persistente nella Valle del fiume Sacco che richiede a tutt’oggi un continuo monitoraggio ed efficaci interventi di bonifica per il recupero ed il risanamento del territorio. Un secondo obiettivo di ricerca ha riguardato la determinazione analitica degli HCH in campioni di pioppo, una pianta arborea utilizzata per il fitorimedio nella Valle del Sacco, per verificarne l’effettiva capacità di assorbimento degli inquinanti. A tale scopo è stato messo a punto un innovativo approccio di tipo dendrochimico tramite una procedura analitica che si è avvalsa, in via preliminare, della spettroscopia IR. Questa parte della ricerca ha richiesto carotaggi di pioppi collocati a diverse distanze dal fiume Sacco, l’analisi degli standard e dei campioni tal quali tramite spettroscopia IR e la successiva validazione dei dati acquisiti tramite gas-cromatografia. | it |
dc.description.abstract | The research carried out during the PhD course is part of an interdisciplinary project funded by INAIL (Istituto Nazionale per l’Assicurazione contro gli Infortuni sul Lavoro) named ”Study of the contamination by hexachlorocyclohexane isomers in the Sacco’s River Valley and evaluation of the risks for the workers of this area”. The Sacco’s River Valley is a wide territory located between Rome and Frosinone in the central Italy crossed by Sacco River. Since 1900, this area had an important industrial development, mainly thanks to Bomprini Parodi Delfino (B.p.d), a company which over time has created a city around the industrial agglomerate. During the two World Wars, the B.d.p company produced mostly military explosives such as nitric acid, glycerine and dynamite. After the Wars , it expanded its range of manufacturing to the chemical industry. In particular, it specialized in synthesizing pesticides, insecticides, fungicides and herbicides and different types of fertilizers. Nevertheless, the intense industrial activity, the absence of laws in waste management and the uncontrolled use of the river water has led to a remarkable pollution of the river, groundwater and soil causing serious problems to the whole agro-food chain. The focus on this emergency has occurred in 2005, after the detection of high levels of toxic substances to human into several sample of milk produced from local farms (up to 20 times higher than the legal limit). The contamination was referred to b- hexachlorocyclohexane (b-HCH), a pesticide belonging to family of organochlorine pesticides produced from B.d.p. and widely used in agriculture. This substance is an halogenated compound belonging to a family of several stereoisomers, named a-, b-, g-, d-, e-, h-, q which differ for the physical-chemical properties and persistence in the environment related to the axial-equatorial substitution pattern of chlorine atoms around the six carbons ring [1]. Among the isomers, the g-HCH, also known as lindane, showed the highest pesticidal activity and it was widely used as an insecticide on fruits and vegetable, rice paddies, animals and seed treatment. Starting of these literature data, the research carried out during the PhD was focused on the investigation of the occurrence and distribution of HCHs isomers in different samples (soil, water, milk, feed and vegetables) collected in nine dairy farms located in the Sacco’s River Valley. The samples were collected in June-September 2013 and January-April 2014. After the appropriate preparation, all samples were analyzed by GC-ECD and GC-MS. At several years from the onset of the agroenvironmental crisis, our analytical data confirmed a high degree of pollution in the investigated area, which deserves continuous environmental monitoring due to the strong impact of persistent organic pesticides on rural environment and human food chain. In addition to the monitoring, the bioremediation of the Valley must be seriously taken in account. Among the possible solutions, phytoremediation is applied in the Sacco’s River Valley. It is based on the ability of certain plants to degrade, assimilate, and metabolize pollutants from soils accumulating them or their metabolites in different tissues and organs. In the final part of the PhD course, a preliminary study focused on the fate of HCHs on poplars. Then, in collaboration with forest researchers, several cores were collected from poplar trees selected according to a gradient of HCHs contamination into the Sacco’s River Valley: one poplar located at a distance of about 500 m from the left bank of the Sacco River; a second poplar located close to the river bank and was thus subject to direct contamination. As a blank, several cores were also extracted from a control poplar growing close to a river in an uncontaminated area near Viterbo. All samples were firstly analyzed by Micro-Fourier Transform Infrared (micro-FTIR) Spectrometry and then by GC-ECD, after an appropriate preparation. This preliminary study evidenced two important aspects: the novelty in the analytical method using Micro-Fourier Transform Infrared (micro-FTIR) Spectrometry, a fast and non-destructive technique for the screening of samples; the ability of poplars to absorb HCHs isomers. This result confirms the role of poplars in bioremediation but opens novel questions about the final uses of them. | it |
