Abstract: 
The study is organized into two main topics: a) the qualitative/quantitative characterization of natural substances in plant extracts, and b) biocatalytic transformations of natural substances through immobilized enzymes.
Regarding the first topic, the main phenolic components of a large panel of plant extracts, provided by Aboca S.p.a., have been characterized in detail by applying GC-MS, LC-MS and NMR techniques. The knowledge of the structure of bioactive components in plant extracts is an important topic related to cosmetic and pharmaceutical applications, as well as for the certification of quality processes. In fact, the biological activity of vegetable extracts is strictly connected to integrity and complexity of the original chemical composition. In this system, any substances with its specific activity can contribute to total activity in a synergistic way with the others. This work is part of a series of pharmaceutical analysis and controls necessary for the use of a plant extract as phytochemical.
Regarding the second topic, the study of biocatalytic transformation of natural substance is related to the use of enzymatic processes within industries in pharmaceutical, fine chemical, fuel and biosensor applications. Different immobilization procedures have been reported to improve the stability of the enzyme. In this context, novel nano-structured materials are gradually emerging as promising supports for immobilization, mainly due to their high surface area, high loading capacity, poor resistance to mass transfer and favourable physical and chemical properties. The aim of this work is to design and characterize biocatalysts based on carbon nanotubes (CNTs) and to apply such systems in synthetic transformations. The attention was directed to laccase from Trametes versicolor and the immobilization techniques included physical methods, covalent bond linkage and the Layer by Layer technique. In the presence of redox mediators, such as TEMPO, the novel biocatalysts were selective and efficient systems for the synthesis of aldehydes in high yield and under mild experimental conditions, with better results than those previously obtained with other immobilized laccases.

Lo studio è organizzato in due argomenti principali: a) caratterizzazione qualitativa/quantitativa di sostanze naturali in estratti vegetali, e b) trasformazione biocatalitica di sostanze naturali attraverso enzimi immobilizzati.
Riguardo il primo argomento, i composti fenolici principali di estratti vegetali, forniti dalla società Aboca S.p.a., sono stati caratterizzati in dettaglio attraverso le tecniche di GC-MS, LC-MS e NMR. La conoscenza della struttura dei componenti bioattivi negli estratti vegetali è un’importante tema collegato ad applicazioni cosmetiche e farmaceutiche, come per la certificazione dei processi di qualità. Infatti, l’attività biologica egli estratti vegetali è strettamente connessa all’integrità e complessità della composizione chimica originale. In questo sistema, ogni sostanza con la sua specifica attività può contribuire all’attività totale in modo sinergico con le altre. Questo lavoro è parte di una serie di analisi farmaceutiche e controlli necessari per l’uso di estratti vegetali come fitofarmaci.
Riguardo il secondo argomento, lo studio delle trasformazioni biocatalitiche di sostanze naturali è relativo all’uso di processi enzimatici nelle industrie farmaceutiche, chimiche, dei carburanti e applicazioni in biosensoristica. Sono state riportate differenti procedure di immobilizzazione per migliorare la stabilità dell’enzima. In questo contesto, nuovi materiali nano-strutturati stanno gradualmente emergendo come promettenti supporti per l’immobilizzazione, principalmente per la grande area di superficie, alta capacità di carico, scarsa resistenza al trasferimento di massa e proprietà fisiche e chimiche favorevoli. Lo scopo di questo lavoro è progettare e caratterizzare biocatalizzatori basati su nanotubi di carbonio (CNTs) e applicare tali sistemi in trasformazioni sintetiche. L’attenzione è stata diretta alla laccasi da Trametes versicolor e le tecniche di immobilizzazione includono metodi fisici, legami covalenti e tecnica Layer by Layer. In presenza di mediatori redox, come il TEMPO, i nuovi biocatalizzatori sono sistemi selettivi ed efficienti per la sintesi di aldeidi con un’alta resa e sotto condizioni sperimentali blande, con miglior risultati di quelli precedentemente ottenuti con altre laccasi immobilizzate.
