Long and complex geological history and long-term favorable climate of the Mediterranean offer a vast number of particular microhabitats inhabited by isolated, highly specialized species and subspecies. Around 10% of all the existing plant species in the world inhabit this small area, which offers always new discoveries and particularities. High genetic richness appears to be particularity of the area and is still in the state of its recognition and investigation. In this thesis, four Mediterranean species/species complexes (European Black Pine, Aleppo Pine, European Nettle Tree and Inula verbascifolia group) were selected to explore their evolutionary relationships. The aim of this study was to provide new insights about systematics and phylogeography of the studied species. Besides, different types of DNA markers and phylogenetic methods for their applicability and usefulness in taxonomic research were evaluated. Finally the comparative genetic variation of four Mediterranean taxa differing in their ecology, biology, life-history traits and distribution was assessed. This study has demonstrated big genetic variability among the species inhabiting the Mediterranean Basin. The great genetic variability between the Aleppo Pine individuals has been found in the eastern populations, especially on the Balkan Peninsula, which appears to be ancestral, based on these analyses. European Black Pine showed the genetic variability between the populations on the Apennine Peninsula, region especially rich of refugia. These populations should be re-analyzed in detail, conserved and protected from artificial importation of the foreign genetic material. Further on south, on the Sicily Island, there has been found particular identity of Celtis tourniforti (informally called Celtis aethnensis), which also demonstrated its genetic variability. Another region of great genetic variability was demonstrated to be Adriatic Sea, especially northern Adriatic coast which, on its numerous islands and islets, offers the vast number of genetically unique populations. Ancient relationships between the opposite Adriatic coasts have been recorded among the Inula verbascifolia subsp. verbascifolia samples. In the study there has been tested the use of the markers from a big number of DNA regions which can provide a comprehensive overview of the methodology recommended for the use for the future analyses on these species. Finally, for the first time high resolution melting (HRM) analyses application has been tested on such a study (study on genetic variability, systematics and phylogeography). Il Bacino Mediterraneo è una regione geografica nota per la sua elevata biodiversità. Grazie alla sua lunga e complessa storia geologica, e ad un clima favorevole, da lungo tempo il Bacino ospita un gran numero di microhabitat, popolati da specie e sottospecie isolate e che nel tempo si sono particolarmente adattate. Circa il 10% della flora fino ad ora conosciuta al mondo, abita questa piccola zona che offre alla comunità scientifica sempre nuove scoperte. La sua unicità é data anche dalla grande ricchezza genetica, la quale rappresenta un’altra particolarità. Nel presente progetto tesi, sono state selezionate quattro specie (pino nero, pino d'Aleppo, bagolaro e Inula verbascifolia gruppo) al fine di studiarne le loro storia evolutiva. Il principale obiettivo di questa ricerca è stato l’approfondimento delle conoscenze di sistematica e filogeografia delle specie in questione. In’oltre sono stati analizzati vari tipi di marcatori molecolari e metodi filogenetici per valutarne la loro applicabilità e utilità nelle ricerche tassonomiche. Infine, è stata condotta un’analisi comparativa della variabilità genetica tra le quattro specie mediterranee le quali si differenziano tra loro nella distribuzione, biologia ed ecologia. I risultati del presente studio hanno dimostrato che nel Bacino Mediterraneo esiste una grande variabilità genetica intra-specie. La variabilità del pino d’Aleppo é stata trovata all’interno dei popolamenti orientali, specialmente sulla penisola Balcanica, regione geografica che potrebbe essere considerata il centro della distribuzione di questa specie. Il pino nero ha dimostrato una variabilità tra i popolamenti presenti sulla penisola Appenninica, area particolarmente ricca di rifugi glaciali. Per questa ragione questi popolamenti dovrebbero essere oggetto di studi più dettagliati mirati a promuovere la loro conservazione e proteggere la loro variabilità genetica, salvaguardandoli dall’introduzione di genotipi esterni. In Sicilia, a sud del tratto appenninico, è stato segnalato un altro caso di variabilità genetica, relativo alla sottospecie Celtis tourniforti (chiamata anche Celtis aethnensis). Un’altra regione riconosciuta per la sua ricchezza genetica è il Mar Adriatico, in particolar modo la sua costa settentrionale che ospita nelle sue numerose isole un numero considerevole di popolamenti geneticamente unici. Nella presente tesi é stata inoltre segnalata un’ulteriore variabilità intraspecifica tra le coste opposte del mare Adriatico, che segnala l’esistenza di antiche relazioni tra gli individui della specie Inula verbascifolia subsp. verbascifolia. In questo studio sono state utilizzate diverse regioni di DNA come marcatori molecolari che possono risultare utili ed informative per ulteriori ricerche future sulle specie oggetto di studio. Infine, é stata testata la metodologia del high resolution melting (HRM) applicata in questa ricerca che comprende in modo combinato studi di variabilità genetica, sistematica e filogeografia. |
