Using these assays we have identified a number of novel substrates of the SCFβTrCP and the APC/CCdh1 ubiquitin ligases. We have made the decision to focus on three transcriptional regulators, namely TFAP4, targeted for degradation by SCFβTrCP, and E2F3 and E2F6, targeted for degradation by APC/CCdh1. We have found that proteasomedependent degradation of these three transcriptional regulators occurs during cell cycle exit and neuronal differentiation. The major aims of this work are to biochemically characterize the proteasome-dependent degradation of TFAP4, mediated by SCFβTrCP, and the ones of E2F3 and E2F6, mediated by APC/CCdh1, and to investigate the biological function of their degradation during cell cycle exit and neuronal differentiation. Abbiamo messo a punto vari saggi basati su strategie di immunopurificazione seguite da analisi di spettrometria di massa per identificare nuovi substrati delle ubiquitina ligasi APC/C e SCF. Utilizzando questi saggi, abbiamo identificato una serie di nuovi substrati delle ubiquitina ligase SCFβTrCP e APC/CCdh1. Abbiamo deciso di focalizzare la nostra attenzione su tre regolatori trascrizionali: TFAP4, ubiquitinato da SCFβTrCP, e E2F3 e E2F6 ubiquitinati da APC/CCdh1. Abbiamo scoperto che la degradazione proteosoma-dipendente di questi tre regolatori trascrizionali avviene durante l’uscita dal cicle cellulare e durante il differenziamento neuronale. Gli obiettivi principali di questo studio sono la caratterizzazione biochimica della degradazione proteosoma-dipendente di TFAP4 mediata da SCFβTrCP, e quella di E2F3 e E2F6 mediata da APC/CCdh1, e l’analisi della funzione biologica della loro degradazione durante l’uscita dal ciclo cellulare e durante il differenziamento neuronale |
