This work focused on the development and the scale up of processes for the production of enzymes from lignocelluloses by Solid State Fermentation (SSF) using a novel prototype demonstration plant at pre-industrial scale. The optimization of thesemprocesses, the study of new products and their potential for “green” applications are also essential parts of the present work. Industrial productions are characterized by a deep and close correlation among the product, the production process and the plant. The contemporaneous development and the strict balance of those three aspects is the main basis for the success of any technologically novel rising industrial activity. As it results from the data of the scientific literature, in order to obtain profitable results it is necessary to develop specific plants and dedicated bioreactors which can take into account some particular characteristics of the new processes such as dissipation of heat due to the metabolic respiration and/or hyphen fragility if filamentous fungi are used. The high flexibility and the low utilities requirement are the most valuable qualities of the plant design obtained. We performed most of the experiments using edible mushrooms strains (mainly Lentinula edodes and Pleurotus ostreatus strains) setting the major interest on the production of extracts containing a pool of enzymes, almost entirely oxidoreductases. A considerable part of this study was devoted to the investigation of novel applications based upon these enzymes, focusing especially on laccase which is highly promising for environmental features. The results of the experiments shown the concrete possibility to apply laccases to the biodeinking treatment of wastepapers and suggest that ligninolytic enzymes can be useful utilized in novel remediation processes for recalcitrant pollutants removal. In accordo con il moderno ed oggigiorno comune concetto di sostenibilità, le biomasse suscitano crescente interesse sia come fonti di energia sia come valida materia prima per la produzione di un gran numero di composti, prodotti chimici e beni di consumo. Questo lavoro si incentra sullo sviluppo e lo scale up di processi per la produzione di enzimi da lignocellulose tramite Fermentazioni in stato solido (SSF) utilizzando un innovativo impianto pilota dimostrativo su scala pre-industriale. L’ottimizzazione di questi processi, lo studio di nuovi prodotti e del loro potenziale in applicazioni “verdi” sono altre parti essenziali del presente elaborato. Le produzioni industriali sono caratterizzate da un’ intima correlazione tra prodotto, processo produttivo ed impianto di produzione. Lo sviluppo simultaneo e lo stretto equilibrio di questi tre aspetti costituisce la base fondamentale del successo di ogni nascente attività industriale innovativa. Come risulta dai dati della letteratura scientifica, per ottenere risultati vantaggiosi é necessario lo sviluppo di impianti specifici e di bioreattori dedicati che tengano conto di alcune particolari caratteristiche dei nuovi processi come la dissipazione del calore dovuto alla respirazione metabolica e/o la fragilità delle ife se vengono usati funghi filamentosi. L’elevata flessibilità e lo scarso consumo delle utility sono le caratteristiche di maggior valore del design d’impianto ottenuto. Abbiamo condotto la maggior parte degli esperimenti utilizzando ceppi di funghi eduli (principalmente Lentinula edodes e Pleurotus ostreatus) volgendo l’interesse maggiormente sulla produzione di estratti contenenti un consorzio enzimatico, costituito prevalentemente da ossidoreduttasi. Una parte considerevole di questo studio è stata dedicata all’investigazione di applicazioni innovative basate su questi enzimi, soffermandosi particolarmente sulla laccasi in quanto altamente promettente per scopi ambientali. |
