Abstract: 
Il glutine è un complesso proteico che conferisce uniche proprietà visco-elastiche agli impasti di frumento ed è di notevole importanza nel determinare la qualità del prodotto finito. Il glutine è composto da due classi proteiche, denominate gliadine e glutenine; le gliadine sono proteine monomeriche in cui le cisteine, qualora presenti, formano esclusivamente legami disolfuro intra-molecolari e sono state ulteriormente classificate in α/β, γ e ω-gliadine in base alla loro mobilità in elettroforesi acida (A-PAGE). Le glutenine sono invece polimeri le cui subunità sono stabilizzate da legami disolfuro inter-molecolari ed, in seguito a trattamento con un agente riducente, rilasciano due tipi di subunità, rispettivamente ad alto (HMW-GS) ed a basso peso molecolare (LMW-GS). Entrambi i gruppi di proteine sono determinanti nel definire le proprietà visco-elastiche degli impasti e loro differenze qualitative e quantitative contribuiscono a determinare le diverse caratteristiche qualitative intervarietali. Le LMW-GS sono state classificate in tre gruppi, B, C e D, sulla base della loro mobilità elettroforetica e del loro punto isoelettrico e, fino ad ora, sono state molto meno studiate e caratterizzate rispetto alle HMW-GS, a causa fondamentalmente del loro elevato numero ed eterogeneità di struttura. Le LMW-GS includono polipeptidi con strutture tipiche (gruppo B) e polipeptidi che sono strutturalmente gliadine ma funzionalmente glutenine (gruppi C e D). Riguardo le subunità di tipo B, esse sono state ulteriormente classificate in LMW-s, LMW-m e LMW-i sulla base della natura del primo amminoacido della sequenza polipeptidica matura, che è rispettivamente serina, metionina ed isoleucina. Mentre le subunità di tipo s e m sono molto abbondanti nell’endosperma di frumento, antecedentemente al presente lavoro di tesi non vi erano dirette e chiare indicazioni circa l’espressione in vivo delle LMW-i.
Un altro aspetto non chiaro riguardante le LMW-GS, è costituito dal processamento N-terminale a cui le LMW-m e le LMW-s sono verosimilmente soggette. Infatti il confronto delle sequenze N-terminali delle LMW-s e delle LMW-m, rivela che esse differiscono principalmente per l’assenza del peptide MET nelle sequenze polipeptidiche mature delle LMW-s. Secondo una recente ipotesi, tale differenza sarebbe causata dal taglio del peptide MEN nelle sequenze di tipo s, operato da un’asparaginil endoproteasi in corrispondenza dell’asparagina localizzata in posizione 23 (N del peptide MEN).
Sulla base di tali premesse, il lavoro sperimentale eseguito per lo svolgimento della presente tesi di Dottorato è stato finalizzato a chiarire alcuni aspetti riguardanti le relazioni struttura-funzione di alcuni membri della complessa famiglia genica delle LMW-GS. In particolare, tramite un approccio integrato basato su tecniche di espressione eterologa, elettroforesi bidimensionale, sequenziamento N-terminale, RP-HPLC e spettrometria di massa MALDI-TOF, è stata dimostrata l’espressione in vivo di una LMW-i e di una γ-gliadina modificata nel numero di cisteine, verosimilmente corrispondente a una LMW-GS di tipo C. Inoltre, le proprietà funzionali dei due polipeptidi sono state studiate attraverso test di incorporazione, eseguiti utilizzando un micro-mixografo prototipo presso il laboratorio di “Dough Rheology” della divisione Plant Industry del centro di ricerche CSIRO (Canberra, Australia). Questi test sono stati condotti sia utilizzando la farina di frumento tenero sia la semola di frumento duro e hanno permesso di dedurre i ruoli funzionali dei due polipeptidi ed, in particolare, il loro contributo nel definire le proprietà visco-elastiche degli impasti.
Un ulteriore aspetto che è stato analizzato nella presente tesi di Dottorato concerne il processamento N-terminale a cui le LMW-s e le LMW-m sono verosimilmente soggette. Al fine di verificare la correttezza dell’ipotesi di un diverso processamento N-terminale, caratterizzato dal taglio del peptide MEN nelle sequenze di tipo s, sono state espresse in Nicotiana benthamiana una LMW-m (LMW-B1133) ed una LMW-s (LMW-42K), wild type e mutate nello specifico residuo in posizione 23 (B1133T23N, sostituzione della treonina in posizione 23 con un’asparagina; LMW-42KN23T, sostituzione dell’ asparagina in posizione 23 con una treonina). Attraverso analisi di Western blotting è stato dimostrato che tutte le proteine ricombinanti (wild type e mutate, con e senza la coda di istidine C-terminale) sono espresse nel tessuto fogliare di Nicotiana benthamiana, sebbene a scarsi livelli. Inoltre, dati preliminari ottenuti dall’analisi di sequenziamento N-terminale della LMW-B1133 wild type e mutata indicano che verosimilmente il peptide MEN è rimosso dalla sequenza matura del polipeptide LMW-B1133T23N anche se, a causa dell’incompletezza dei dati di sequenza ottenuti, non possiamo confermare questo dato con certezza. Questo lavoro è ancora in fieri e, attualmente, stiamo purificando le LMW-GS eterologhe dal tessuto fogliare di tabacco al fine di ottenerne una quantità sufficiente necessaria all’analisi di sequenziamento.

Gluten is a protein complex that confers unique visco-elastic properties to wheat doughs and is of critical importance in determining end product quality. Gluten is composed by gliadins and glutenins. Gliadins are monomeric proteins in which the cysteines, if present, form only intra-molecular disulphide bonds and are classified into α/β, γ and ω-gliadins according to their mobility in lactic acid PAGE (A-PAGE). Conversely glutenins are polymers whose subunits are joined together by disulphide bonds and, after chemical reduction, release high and low molecular weight glutenin subunits (HMW-GS and LMW-GS, respectively). Both groups of proteins are important for the final visco-elastic properties of dough and both qualitative and quantitative differences within these groups of proteins contribute to inter-cultivar variation in quality. LMW-GS are classified into three groups, B, C and D, on the basis of their electrophoretic mobility and isoelectric point and are much less characterized than high molecular weight glutenin subunits, due to their great number and structure heterogeneity. They include polypeptides with peculiar sequence (B group) along with polypeptides that are structurally gliadins (C and D group) but functionally glutenins. Regarding the B subunits, they have been further subdivided into LMW-s, LMW-m and LMW-i types whose mature polypeptides start with Ser, Met and Ile, respectively. Whereas LMW-m and LMW-s are the most common, there were no direct indication about the in vivo expression of LMW-i in wheat endosperm. Another aspect that need further attention is about the N-terminal processing of LMW-m and LMW-s types. In fact the comparison of LMW-m and LMW-s N-terminal sequences reveals that they differ mainly for the absence of the short peptide MET in the LMW-s. Recently it has been hypothesised that the lack of the peptide MET in LMW-s type sequences is likely due to a proteolitic cleavage performed by an asparagynil endoprotease that recognizes the Asn residue in position 23 of the full polypeptide sequence.
On the basis of these preliminary remarks, the experimental work carried out in the present thesis was aimed at clarify some aspects regarding structure-function relationships of some members of the complex LMW-GS family. In particular the in vivo expression of a LMW-i type subunit and of a modified γ-gliadin with an additional cysteine located at the beginning of the repetitive domain was demonstrated by means of an integrated approach based on two-dimensional electrophoresis, N-terminal sequencing, RP-HPLC, MALDI-TOF mass spectrometry. The functional properties of the two polypeptides were indeed investigated by means of incorporation tests carried out by using a prototype 2-g Mixograph at the “Dough Rheology” laboratory, Plant Industry division, CSIRO (Canberra, Australia). These tests were performed using both bread flour and durum wheat semolina and have allowed us to drawn functional properties exerted by the two polypeptides.
Another aspect that has been analysed in the present thesis is the N-terminal processing to which the LMW-s and LMW-m type are likely submitted. In order to confirm the hypothesis of a differential N-terminal processing for the LMW-s and LMW-m type, characterized by the cleavage of the short peptide MEN in the LMW-s sequences, a LMW-m (LMW-B1133) and a LMW-s (LMW-42K), wild type and mutated at position 23 (B1133T23N, substitution of the Thr 23 with an Asn; LMW-42K N23T, substitution of the Asn 23 with a Thr), have been expressed in Nicotiana benthamiana plants by means of a transient heterologous expression system based on potato virus X (PVX). By means of Western blotting analysis it has been clearly shown that all the recombinant proteins (wild type and mutated, with and without the C-terminal his-tag) are expressed in Nicotianana benthamiana leafs, even at low level. Moreover the data obtained from the N-terminal sequencing of the recombinant wild type and mutated LMW-B1133 have shown that the peptide MEN is likely cleaved from the mature polypeptide LMW-B1133T23N. Unfortunately we are not able to confirm definitely the hypothesis of a differential N-terminal processing for the LMW-m and LMW-s, because the N-terminal sequencing data obtained are incomplete probably due to the scarcity of the polypetides sequenced. This work is still in progress and we are currently purifying the recombinant proteins from tobacco leafs in order to obtain the sufficient amount needed to perform a N-terminal sequencing analysis.
