Abstract
My Ph.D. work has been focused on the development of new flow cytogenetic techniques for chromosome manipulation and gene localization in chickpea. Most of the research efforts have been made to set up a proper methodology for celi cycle synchronization and chromosome isolation in suspension best suited for two Italian genotypes. Flow karyotyping from both types desi and kabuli chickpea was achieved using laser excited DNA specific fluorochromes. On DNA histograms, weli defined regions corresponding to single type chromosomes were identified and sorted. Sorted fractions were used for physical mapping of specific sequence linked to Fusarium oxysporum resistance in desi and kabuli chickpea, which was the main goal ofthis Ph.D. Thesis.
From my research work I do hope will be possible to provide the basic methodologies to build subgenomic libraries in chickpea, useful for the identification and physical mapping of regions coding for important agronomic traits, fundamental for the genetic improvement ofthe species, as it was possible to do to other plant species.
