Abstract
Damage to the plant cell wall (CW) is perceived by the plant as a danger signal, leading to the activation of defence mechanisms in which the ARABIDOPSIS NPK1-RELATED PROTEIN KINASES 1-3 (ANP1-3), a family of the triple MAP kinases (MAP3K), are involved. The lack of ANP reduces the cellulose content of the CW, resulting in alterations of the root phenotype similar to those induced by treatment with Isoxaben (ISX), a chemical inhibitor of cellulose biosynthesis, and by perturbation of the functionality of the endoplasmic reticulum (ER). In this work, I studied the molecular mechanisms potentially involved in CW alterations related to ER stress, ISX treatment and to altered ANP expression, discovering the activation of common pathways regulated by JA. My results attribute for the first time in plants an active role of MAP3K in the regulation of ER functionality and related signalling, also showing the involvement of ER stress as integral part of the intracellular response to Cell Wall-Damage Associated Molecular Patterns (CW-DAMPs) such as oligogalacturonides (OGs). OGs and ER stress signalling converge on the same intracellular mechanisms mediated by the subcellular translocation of ANP1 and ANP3. Furthermore, co-immunoprecipitation analysis highlighted among the possible interactors of ANPs proteins resident in the ER and in the related ER bodies. Although further investigations are necessary, my work attributes a new role to ANPs and identifies a new intracellular event involved in the response to OGs that was previously unknown.
Il danneggiamento della parete cellulare vegetale (CW) è percepito dalla pianta come segnale di pericolo, portando all’attivazione di meccanismi di protezione in cui sono coinvolte le triple MITOGEN-ACTIVATED PROTEIN chinasi (MAP3K) denominate ARABIDOPSIS NPK1-RELATED PROTEIN KINASES 1-3 (ANP1-3). La mancanza di ANP riduce il contenuto di cellulosa della CW, determinando alterazioni fenotipiche radicali simili a quelle indotte dal trattamento con Isoxaben (ISX), un inibitore chimico della biosintesi della cellulosa, e dalla perturbazione della funzionalità del reticolo endoplasmatico (ER). In questo lavoro, ho studiato i potenziali meccanismi molecolari coinvolti nelle alterazioni di parete legate all’ER stress, al trattamento con ISX all'alterata espressione di ANP, scoprendo l'attivazione di percorsi comuni regolati da JA. I miei risultati attribuiscono per la prima volta in pianta un ruolo attivo delle MAP3K nella corretta regolazione della funzionalità del ER e della relativa segnalazione che si è dimostrata parte integrante anche nella risposta intracellulare che segue la percezione dei Pattern Molecolari Associati al Danno prodotti dalla parete cellulare (CW-DAMP) come gli oligogalatturonidi (OG). Il trattamento con CW-DAMP e la risposta allo stress del ER confluiscono sugli stessi meccanismi intracellulari mediati dalla traslocazione subcellulare di ANP1 e ANP3. Inoltre, l’analisi di co-immunoprecipitazione ha evidenziato tra i possibili interattori di ANP1 e ANP3 alcune proteine residenti nell’ER e nei derivanti ER bodies. Nonostante siano necessari ulteriori approfondimenti, il mio lavoro attribuisce un nuovo ruolo alle ANPs e ha permesso di identificare un nuovo evento intracellulare coinvolto nella risposta agli OG finora sconosciuto.
