Functional and comparative genome analysis of microorganisms suitable for the developing of microbe assisted phytotechnologies

Abstract
Microbial assisted phytoremediation is a promising remediation technique, which exploit the utilization of plant associated microorganisms for the rehabilitation of polluted environments. The aim of this study was to dissect the biotechnological potential of Rhodotorula graminis WP1 and Rhodococcus atherivorans BCP1 as candidate microorganisms for the developing of a microbial consortium capable to couple important aspects of microbe assisted phytotechnologies (e.g. improved growth performances and increased plant tolerance to environmental stresses) to phytoremediaton. In this context, a comparative genome analysis was performed on WP1 to highlight specific genetic features associated with plant-microbe interactions, and characterize the presence of virulence traits typical of the Pucciniomycotina phytopathogens. Furthermore, from the perspective phytotechnologies, the genetic potential of WP1 involved in the cellular homeostasis of heavy metals, mainly arsenic, was analyzed. Finally, different aspects regarding the homeostasis and response of BCP1 to As(V) and As(III) were investigated using both physiologic and bioinformatic approaches.
Il fitorimedio assistito da microorganismi è una promettente tecnica di bonifica che sfrutta l'utilizzo di microrganismi associati alle piante per il recupero di ambienti inquinati. Lo scopo di questo lavoro è stato quello di studiare il potenziale biotecnologico di Rhodotorula graminis WP1 e Rhodococcus atherivorans BCP1 come microrganismi candidati per lo sviluppo di un consorzio microbico in grado di accoppiare importanti aspetti delle fitotecnologie microbiche (es. migliori prestazioni di crescita e maggiore tolleranza delle piante agli stress ambientali) al fitorimedio. In questo contesto, è stata eseguita un'analisi comparativa del genoma di WP1 per evidenziare specifiche caratteristiche genetiche associate alle interazioni pianta-microbo e caratterizzare la presenza di tratti di virulenza tipici dei fitopatogeni facenti parte del gruppo tassonomico Pucciniomicotina. Inoltre, sempre dal punto di vista delle fitotecnologie, è stato analizzato il potenziale genetico di WP1 coinvolto nell'omeostasi cellulare di metalli pesanti, principalmente arsenico. Infine, diversi aspetti riguardanti l'omeostasi e la risposta di BCP1 a As (V) e As (III) sono stati studiati utilizzando approcci sia fisiologici che bioinformatici.
