Abstract
This thesis was carried out in order to contribute to enlarge knowledge on the biological properties of Aloe arborescens plant. MTT assays showed an intense antiproliferative activity of Aloe arborescens leaf skin extracts tested on murine myeloma cells (P3X) and human promyelocytic cells (HL60). Cytological study have been carried out with immunofluorescence techniques and with SEM and TEM observations. Chemical analysis of the leaf extract with TLC, HPLC and LC-mass investigations showed the presence of the molecules aloenin and the aloins A and B, 2’O-feruloylaloesin. The spot E, isolated by TLC, showing antiproliferative activity and NMR investigations showed that this spot consisted of the molecules aloenin and the aloins A and B. The concentration of single molecules was confirmed by HPLC investigations and single molecules and solutions formed by different combination of single molecules were tested on P3X and HL60. Since after administration of few concentration for long time of Aloe arborescens leaf extract some murine myeloma cells survived, attention was focussed to the surviving cells and related induction of drug-resistance properties; such cells showed an increasing of cell volume and were able to proliferate. In order to evaluate a possible molecular marker, the expression of PGP and p53 proteins ware tested by western blot techniques and immunofluorescence investigations. Further, control cells untreated with Aloe arborescens leaf extract were then cultured in culture medium of resistant cells and drug resistant properties resulted acquired, thus confirming the presence in the culture medium of a drug resistance inducing factor.
