Abstract
The end of the "era" of synthetic drugs and the development of techniques and technologies of investigations based on computational methods, such as High Content Analysis (HCA), have allowed natural products (NPs) to come back as protagonists in the drug discovery scenery. Autophagy, a physiological cellular process for the maintenance of the cellular homeostasis, is currently one of the most investigated cellular process because involved in many common diseases of the modern age, for example cancer, neurodegeneration and inflammation. In this work an analytical approach based on High Content Analysis methods for the investigation of the effects of NPs on autophagy is presented.
The effects of forty-seven extracts from different organisms, like algae, invertebrate and plants, have been investigated by setting up fluorescence and immunofluorescence assays based on autophagy markers; quantitative data describing autophagic phenotypes obtained from the resulting images were then used to define qualitatively their effects on autophagy.
La fine dell'"era" delle droghe sintetiche e lo sviluppo di tecniche e tecnologie di investigazione basate su metodi computazionali, come l’High Content Analysis (HCA), hanno garantito un ritorno dei prodotti naturali (NP) come protagonisti nel panorama della ricerca di nuovi farmaci. L'autofagia, un processo cellulare fisiologico per il mantenimento dell'omeostasi cellulare, è attualmente uno dei processi cellulari più studiati perché coinvolto in molte malattie comuni dell'età moderna, come ad esempio il cancro, la neurodegenerazione e l'infiammazione.
In questo lavoro viene presentato un approccio analitico basato su metodi di l’High Content Analysis per lo studio degli effetti dei NP sull'autofagia. Gli effetti di quarantasette estratti di diversi organismi, come alghe, invertebrati e piante, sono stati studiati attraverso l’utilizzo di saggi di fluorescenza e immunofluorescenza basati sull’osservazione di marcatori autofagici; i dati quantitativi relativi ai fenotipi autofagici ottenuti dalle immagini risultanti sono stati poi utilizzati per definire qualitativamente i loro effetti sull'autofagia attraverso le tecniche di High Content Analysis.
