Abstract
The management and disposal of olive mill waste-water (OMWW) are among the main environmental problems of the Mediterranean area because the phenolic nature of OMWW makes it highly resistant to biodegradation and highly phytotoxic. However, in the OMWW many high addedvalue compounds can be recovered to use in other ways. Among these, Hydroxytyrosol (HTyr) is a phenolic compound with various beneficial effects on human health and it can cross the blood-brainbarrier. Considering the previous data on neuroprotective effects of HTyr, we asked whether HTyr could stimulate hippocampal neurogenesis in vivo in adult and aged wild-type mice as well as in the Btg1 knockout, a mouse model of accelerated neural aging. We found that treatment with HTyr activates neurogenesis in the dentate gyrus of adult, aged and Btg1-null mice, by increasing the survival of new neurons and decreasing apoptosis. Notably, however, in the aged and Btg1-null dentate gyrus, HTyr treatment stimulates also the proliferation of stem and progenitor cells, whereas in the adult dentate gyrus HTyr lacks any proliferative effect. Moreover, the new neurons generated in aged mice after HTyr treatment are recruited to existing circuits, as shown by the analysis of specific neural markers. Finally, HTyr treatment also reduces the markers of aging lipofuscin and Iba1. Overall, our findings suggest that the recovery of HTyr from OMWW could be an advantage for the environment, and its use as a diet supplement an important ingredient to counteract neurogenesis decline during aging.
La gestione e lo smaltimento delle acque di scarto prodotte durante la lavorazione delle olive (OMWW, olive mill waste water) è sono tra i principali problemi ambientali dell’area mediterranea. Infatti, l’alta concentrazione fenolica presente in questi rifiuti li rende altamente fitotossici e scarsamente biodegradabili. Tuttavia alcune di queste molecole fenoliche hanno anche un potenziale benefico sulla salute dell’organismo, in particolare sono oggetto di studio le proprietà dell’idrossitirosolo (HTyr). Considerando l’attività anti ossidante dell’HTyr e la sua capacità di oltrepassare la barriera ematoencefalica, nonché i già noti effetti neuroprotettivi dell’HTyr, ci siamo chiesti se l’HTyr potesse essere utilizzato come stimolo neurogenico. A tal fine, abbiamo testato le capacità neurogeniche dell’HTyr in topi anziani, adulti e nel modello murino Btg1 knockout, un modello murino di invecchiamento accelerato. I risultati ottenuti hanno dimostrato che il trattamento con l’HTyr attiva promuove la produzione di nuovi neuroni nella nicchia neurogenica del giro dentato dell’ippocampo di topi adulti, anziani e Btg1 knockout, promuovendo la sopravvivenza cellulare e inibendo l’apoptosi. Il trattamento con HTyr è anche in grado di stimolare l’attività proliferativa delle cellule staminali e progenitrici neurali nelle condizioni deficitarie del topo anziano e nel topo adulto Btg1 knockout, mentre tale attività pro-proliferativa è assente nel giro dentato del topo adulto wild type. Inoltre, i nuovi neuroni prodotti nel topo anziano in seguito al trattamento con HTyr sono reclutati nei circuiti neurali preesistenti, come mostrato dall’analisi di specifici marker neurali. Infine, il trattamento con l’HTyr riduce la presenza di lipofuscina e di cellule microgliali attivate nel giro dentato del topo anziano, suggerendo un miglioramento dell’ambiente della nicchia neurogenica. Complessivamente, questi risultati incoraggiano l’utilizzo di HTyr come supplemento dietetico al fine di combattere il declino neurogenico che si osserva con l’invecchiamento o in condizioni neurogeniche deficitarie, incentivandone così il suo recupero dalle OMWW al fine di rendere la produzione dell’olio d’oliva più sostenibile.
