Resistenza in pesco a Xanthomonas arboricola pv. pruni: caratterizzazione della risposta metabolica attraverso analisi differenziale del trascrittoma

Abstract
Background: bacterial spot caused by Xanthomonas arboricola pv. pruni (Xap) is the most dangerous bacterial disease for Prunus species. The peach cv. Redkist is spot resistant while J.H. Hale is spot sensitive. In order to understand the molecular mechanisms underlying the incompatible or compatible interaction of peach and Xap, deep transcriptome sequencing (RNA-Seq) for differential gene expression profiling was performed for both genotypes in a time course analysis 30 min, 1 h, 3h post-infection (hpi).
Results: RNA-Seq analysis detected in peach leaf 20,837 expressed genes representing 75% of peach genome predicted genes. A total of 838 and 803 differentially expressed genes (DEGs) were identified as spot responsive genes in J.H. Hale and Redkist respectively. The former regulated 53.4% of DEGs (448) at 3 hpi while the latter 88.9% (714). Both varieties induced genes involved in signal transduction, hormone biosynthesis, secondary metabolism, ROS burst, and defense proteins, and transcription factors.
Conclusions: this study revealed very similar early molecular responses to bacterial spot disease in Redkist and J.H. Hale after bacterial infection; key differences were detected in the expression of transcription factors, salicylic acid metabolism, ROS burst, shedding light on the resistance response exerted by Redkist. This work provided valuable information for the identification of candidates for engineering spot resistance to obtain resistant varieties.
