Abstract
Negli ultimi anni le regioni polari hanno subito i drammatici effetti del riscaldamento globale. Determinare l'attuale composizione delle comunità microbiche di questi ambienti e definire i parametri ambientali cruciali per la loro diffusione è fondamentale per monitorare i cambiamenti futuri e prevedere i possibili effetti della variazione delle condizioni ambientali. A tal fine, abbiamo utilizzato l'approccio di metabarcoding, che rappresenta un metodo rapido e affidabile per lo studio delle comunità microbiche. Data la crescente diffusione della vegetazione arbustiva riportata in tutto l'Artico, abbiamo studiato le comunità fungine e batteriche associate a tre diversi habitat terrestri, rappresentativi di crescente copertura vegetativa, nella Groenlandia occidentale. Il tipo di copertura era il principale determinante della composizione delle comunità e la composizione della comunità arbustiva un parametro fondamentale nei plot in cui era presente. I risultati hanno mostrato un numero elevato di specie uniche degli ecosistemi più semplici, che potrebbero scomparire con l'espansione degli arbusti, suggerendo una possibile perdita di diversità locale. In Antartide, abbiamo preso in considerazione due diversi ecosistemi tra i più rappresentativi di questo ambiente. In primo luogo, abbiamo analizzato la composizione batterica delle comunità criptoendolitiche dominate da licheni in un gradiente di crescente pressione ambientale. La struttura delle comunità batteriche è risultata essere correlata con la controparte fungina e, tra i parametri ambientali considerati, l'altitudine è risultata essere il principale fattore che influenza la biodiversità batterica. Successivamente, abbiamo analizzato la componente fungina di croste biologiche di suolo presenti lungo le coste della Terra Vittoria durante l'estate australe. I risultati hanno mostrato un maggiore effetto dei fattori abiotici sulla diversità e sulla composizione della comunità. Il forte effetto del pH e della capacità di scambio cationico sulla composizione della comunità fungina ci consente di ipotizzare che in tali ambienti limitanti, anche piccoli cambiamenti nelle condizioni esterne possano influenzare drammaticamente la disponibilità dei nutrienti e quindi l'equilibrio di questi ecosistemi.
In recent years, polar regions have undergone the dramatic effects of global warming. Determining the current composition of the microbial communities of these environments and defining the environmental parameters that are crucial for their spread is fundamental to monitor future changes and predict the possible effects of the variation of environmental conditions. For these purposes, we used the metabarcoding approach, that represents a rapid and reliable method for studying microbial communities. Given the greening and shrub encroachment reported all over the Arctic, we studied both the fungal and bacterial communities associated to three different soil habitats in Western Greenland, representative of a vegetation increasing coverage. The type of coverage was the main predictor of community composition and the shrub composition a fundamental parameter in vegetated plots. The results showed a high number of species unique of the simplest ecosystems, that are expected to be outcompeted with the expansion of shrubs, suggesting a possible loss of local diversity. In Antarctica, we took into account two different ecosystems among the most representatives of this environment. Firstly, we analyzed the bacterial composition of cryptoendolithic lichen-dominated communities in a gradient of increasing environmental pressure; the structure of the bacteria communities showed to be correlated with the fungal counterpart and, among the environmental parameters considered, the altitude was found to be the main factor influencing the bacterial biodiversity. Later, we analyzed the fungal component of biological soil crusts spreading along the Victoria Land coasts during austral summer. The results showed a stronger effect of the abiotic factors on the diversity and community composition. The strong effect of pH and cation exchange capacity on fungal community’s composition let us hypothesize that in such nutrient limiting environments, even small changes in the external conditions can dramatically affect the nutrient availability and thus the balance of these ecosystems
