Applicazione del melasso da barbabietola come substrato alternativo per la crescita e l’accumulo di pigmenti antiossidanti/nutraceutici di Haematococcus puvialis

Abstract
The microalgae offer a large amount of material from which extract useful components. Haematococcus pluvialis is a green single-celled photosynthetic microalgae can accumulate carotenoids with known antioxidant and nutraceutical activities, in addition to antilipidemics and anti-inflammatory activities. The normal growth substrate for in vitro cultures of this organism in the laboratory is the BG11 medium.
In the present study was evaluated the use of beet molasses, the mother liquor remaining after the extraction of sugar from beet juice, as a substrate alternative to the growth of H. pluvialis. Additional parameter for the assessment of the growth of microalgae in this substrate was the extraction and quantification of the carotenoids, focusing on those known for their antioxidant capacity.
Cultures of H. pluvialis in BG11 were established; this culture, as well as comparison with other cultures, was used as stock culture for inocula in medium containing respectively 0.1, 0.3, 0.5, 0.8, 1.0, 1.3, 1.5 and 2.0 g/l of molasses and 1.0 g/l of sodium nitrate and magnesium sulfate heptahydrate, these two of lower purity of laboratory salts. The cultures obtained were monitored for 7 days to obtain the various growth curves; the seventh day was carried out the extraction of the carotenoids.
The HPLC analysis was performed with a reversed phase column and a Shimadzu instrument equipped with autosampler, two pumps and a detection system composed of a DAD (Diode Array Detector) and a in line mass spectrometer, always of Shimadzu. The identification of the carotenoids of interest to us, zeaxanthin, lutein, anteraxanthin, violaxanthin and astaxanthin was made by comparing the retention times and peak areas of analytical standards with the extracts obtained. Were also assessed the costs of using beet molasses on a hypothetical microalgae plant of 1000 liters of growth substrate, on paper, comparing the cost of using a classical mixture of salts from the laboratory and the innovative one with beet molasses.
