Omics analysis of microgravity effects on osteoblastic cells: mitochondrial alterations and cellular regression of the mature phenotype

Abstract
The microgravity affects on human biology, in particular involved the activity of osteoblastic cells. Hence the interest of investigating, from a molecular point of view, the effects of microgravity on these cell cultures, to understand biological processes, cellular components and molecular functions mostly affected by the absence of gravity. In this study, first at all, it’s carried out a preliminary analiysis to investigate eventual differences in cellular vitality between osteoblastic cell grown in microgravity condition and normal condition. This study shows change in mitochondrial activity. This effect was further investigated using proteomic and metabolomic approach. These analyzes show that the absence of gravity induces both a general alteration of mitochondrial proteins and of metabolic pathways connected to the electron transport chain. These alteration could be one of the causes of reduced osteoblastic function. Furthermore, the data obtained from the omic analyses were integrated with histochemical and ultrastructural approaches. By these studies ex vivo it was found that simulated microgravity could trigger a reversible transition to a mesenchymal-like phenotype, triggering a de-differentiation process. In turn allows osteoblastic cells to obtain migrating properties, typical of pre-differentiation stage cells. All this could, in the future, open the doors to innovative therapeutic strategies for the treatment of certain osteo-degenerative pathologies.
La microgravità influisce sulla biologia umana, in particolare sull'attività delle cellule osteoblastiche. Da qui l'interesse di indagare, da un punto di vista molecolare, gli effetti della microgravità su queste colture cellulari, per comprendere processi biologici, componenti cellulari e funzioni molecolari maggiormente interessate dall'assenza di gravità. In questo studio, un’analisi preliminare sulla vitalità cellulare tra cellule osteoblastiche cresciute in condizioni di microgravità e condizioni normali ha permesso di individuare anamalie nell'attività mitocondriale. Questo effetto è stato ulteriormente indagato mediante studi protomici e metabolomici. Queste analisi mostrano che la microgravità induce sia un'alterazione generale delle proteine mitocondriali sia delle vie metaboliche collegate alla catena di trasporto degli elettroni. Ciò potrebbe essere una delle cause della riduzione della funzione osteoblastica. Inoltre, i dati ottenuti dalle analisi omiche sono stati integrati con approcci istochimici e ultrastrutturali. Da questi studi ex vivo è emerso che la microgravità simulata potrebbe innescare una transizione reversibile verso un fenotipo di tipo mesenchimale, attivando un processo di de differenziazione che permette alle cellule osteoblastiche di ottenere proprietà migratorie. Tutto ciò potrebbe, in futuro, aprire le porte a strategie terapeutiche innovative per il trattamento di alcune patologie osteo-degenerative.
