Abstract
Durum wheat (Triticum turgidum ssp. durum) is one of the most important cereal crops worldwide, particularly in the Mediterranean region. In recent years, growing interest has emerged around relatives of durum wheat—such as Khorasan wheat (T. turgidum ssp. turanicum) and Polish wheat (T. turgidum ssp. polonicum)—as well as landraces and historical cultivars from the early 20th century, commonly referred to as conservation varieties. This renewed attention is largely driven by consumers' perception that these traditional wheats produce foods that are healthier and safer than those made from modern cultivars. Additionally, cultivating these ancient wheats reinforces the cultural heritage of regions like Sicily, and they may possess unique agronomic or nutritional traits valuable for breeding programs. In these contexts, it is crucial to explore the differences in kernel composition of these genotypes, through comparative biochemical studies. Moreover, given the economic value of the supply chains of these varieties, it is important to develop methods for varietal identification and traceability. To address these objectives, in this dissertation four different studies were conducted. 1. Comparative Evaluation between durum wheat and its relatives: Various genotypes of durum, Khorasan, and Polish wheat were assessed for kernel accumulation of dietary fiber and minerals, technological quality, and root morphology. The results revealed that observed differences were more genotype-dependent than subspecies-dependent, indicating that broad generalizations across subspecies are not warranted. 2. Gluten Protein Composition in Sicilian Conservation Varieties: The gluten protein profiles of 51 accessions representing 22 Sicilian conservation varieties were analysed to evaluate intra-accession and intra-varietal variability. The electrophoretic patterns of gliadins proved useful in distinguishing between varieties, offering a promising tool for traceability purposes. 3. Multi-Environmental Trial on Five Sicilian Conservation Varieties: Five prominent conservation varieties—Bidì, Russello, Perciasacchi, Timilia reste bianche, and Timilia reste nere—were cultivated across two Sicilian locations over three years, alongside three modern cultivars (Iride, Orizzonte, and Simeto). The study quantified levels of two compounds associated with adverse reactions to wheat: amylase/trypsin inhibitors (ATI) and fructans. The differences in their accumulation were generally minor and more strongly influenced by genotype. 4. Genetic Characterization and Development of a DNA-Based Traceability Method: A total of 141 accessions from the five conservation varieties were genotyped using SNP markers. The resulting data were used to analyse genetic diversity and to develop a robust DNA-based method for varietal identification and traceability. Overall, this dissertation underscores the importance of considering genotypic variation when evaluating traditional versus modern wheats. These findings, along with the traceability methods developed, can contribute significantly to the valorization of conservation varieties and help to fill the knowledge gap on the differences on kernel composition introduced by breeding.
Il frumento duro (Triticum turgidum ssp. durum) è una delle colture cerealicole più importanti a livello mondiale, in particolare nell’area del Mediterraneo. Negli ultimi anni vi è un crescente interesse per alcune specie di frumento tetraploide poco coltivate, come il khorasan (T. turgidum ssp. turanicum) e il polonico (T. turgidum ssp. polonicum), e per le landraces e le cultivar storiche di frumento duro coltivate prima della Rivoluzione Verde, definite come varietà da conservazione. Questa rinnovata attenzione è in gran parte motivata dalla percezione dei consumatori che i prodotti derivanti da queste tipologie di frumento siano più salutari e sicuri rispetto a quelli ottenuti da cultivar moderne. Inoltre, queste varietà hanno un valore culturale importante, soprattutto in Sicilia, e sono interessanti perché potrebbero avere tratti agronomici o nutrizionali unici, utili nei programmi di miglioramento genetico. In questo contesto, è fondamentale esplorare le differenze nella composizione della cariosside di questi genotipi attraverso studi biochimici comparativi. Inoltre, dato il valore economico delle filiere legate a queste varietà, è importante sviluppare metodi per l’identificazione varietale e la tracciabilità. A tal fine, in questa tesi sono stati condotti quattro studi differenti: 1. Analisi comparativa tra frumento duro, khorasan e polonico: Diversi genotipi di frumento duro, khorasan e polonico sono stati valutati per l’accumulo di fibra alimentare e minerali nella cariosside, qualità tecnologica e morfologia radicale. I risultati hanno evidenziato che le differenze osservate dipendono più dal genotipo che dalla sottospecie, indicando che non sono giustificate generalizzazioni troppo ampie tra queste. 2. Composizione delle proteine del glutine delle varietà da conservazione siciliane: Sono stati analizzati i profili proteici del glutine di 51 accessioni rappresentanti 22 varietà da conservazione siciliane, al fine di valutare la variabilità intra-accessione e intra-varietale. I pattern elettroforetici delle gliadine si sono rivelati utili nel distinguere tra varietà, offrendo uno strumento promettente per la tracciabilità. 3. Accumulo di ATI e fruttani in cinque varietà da conservazione siciliane: Cinque importanti varietà da conservazione siciliane, Bidì, Russello, Perciasacchi, Timilia reste bianche e Timilia reste nere, sono state coltivate in due località della Sicilia per tre anni, insieme a tre cultivar moderne (Iride, Orizzonte e Simeto). Sono stati quantificati due composti associati a reazioni avverse al frumento: gli inibitori delle amilasi e della tripsina (ATI) e i fruttani. Le differenze nel loro accumulo sono risultate generalmente modeste e influenzate soprattutto dal genotipo. 4. Caratterizzazione genetica e sviluppo di un metodo di tracciabilità basato sul DNA: Un totale di 141 accessioni delle cinque varietà da conservazione siciliane è stato genotipizzato utilizzando marcatori SNP. I dati ottenuti sono stati utilizzati per analizzare la diversità genetica e sviluppare un metodo solido, basato sul DNA, per l’identificazione varietale e la tracciabilità. Complessivamente, questa tesi sottolinea l’importanza di considerare la variazione genotipica nella valutazione dei frumenti tradizionali rispetto a quelli moderni. I risultati ottenuti, insieme ai metodi di tracciabilità sviluppati, possono contribuire significativamente alla valorizzazione delle varietà da conservazione e fare luce sulle possibili differenze nella composizione della cariosside introdotte dal miglioramento genetico.
