The effect of arbuscular mycorrhiza on micropropagated olive (Olea Europaea L.) plant growth

Abstract
The effect of three strains of arbuscular mycorrhiza fungi (AMF) from Indonesia were
studied on olive plantlets of cv Moraiolo and Canino previously propagated in vitro, both transgenic for rolABC and osmotine genes, and non transgenic. First, the studies
characterized the morphology, the physiology and the molecular aspects of the fungi used for the experiments. Then the stability of transgenes was tested. Results showed that molecular studies did not allow the distinction of the two types of Gigaspora, but it was possible to distinguish them through morphological analysis. Transgenic cultivars of olive (osmotin and rolABC) used for the experiment continued to express the transgene even after several years in vitro. Furthermore, studies were conducted to improve proliferation of cv Canino by using
Dikegulac, a compound that inhibit gibberellins biosynthesis in plants. It was very effective for reducing the apical dominance, and for increasing the number of shoots that could be suitable for the rooting phase. Rooting was improved by exposing the shoots to 16 hours of
light after being kept to 4 days in continuous darkness. Infection with strains of AMF improved the growth of micropropagated non-transgenic plants and their survival rate.
However, growth differences were observed among the strains of AMF used. This
experiment designed using a poor soil in structure, showed better growth difference among seedlings and mycorrhization respect to the control. The reduction of transgenic plants for osmotin to establish symbiosis with AMF strains was not observed even if the could present a fungicidal action of the protein over-expressed in these plants.
