Identification of chromosome 5A encoded polypeptides in wheat kernels

Abstract
This PhD work has been performed in the frame of the Italian project “Physical map of wheat chromosome 5A: Italian initiative for the sequencing of the whole genome”, that is part of the international initiatives ETGI & IWSC. Wheat kernel proteins are represented for 80% of gluten and for 20% by soluble proteins. Whereas chromosome localization of the former is well known, that of soluble proteins, including polypeptides with structural and metabolic functions, still needs to be identified and mapped. A proteomic approach has been performed to identify polypeptides encoded by genes on chromosome 5A. Intervarietal and interspecific chromosome substitution lines of durum and bread wheat have been used, leading to the identification of 48 soluble proteins in the two tetraploid genotypes taken into consideration, and a total of 86 proteins in the 5 bread wheat cultivars analysed. The biological functions of most of the identified polypeptides are: stress/defense; carbohydrate metabolism, protein synthesis/assembly, storage, and a significative portion corresponds to unknown proteins. The knowledge of polypeptides encoded by genes at chromosomes 5A will allow correlating their presence with specific physiological characteristics, along with quality properties.
