Abstract
Several taxa belonging to the genus Acer, not only at national level, are still poorly characterized from a taxonomical point of view.
Despite the applications of molecular biotechnologies, such as DNA sequencing, have now been acquired as an usual analytic tool in identification of animal and vegetal species, there are no previous studies about the genetic characterization of genus Acer both in nationwide and worldwide.
In this context, the technique of DNA barcoding was applied to evaluate this methodology as Acer taxonomic tool and as a complement to studies of population genetics and phylogeny.
Therefore, were selected 3 chloroplast markers (rbcL, rpoC1, trnH-psbA) among those suggested by the Conference of Taipei and the nuclear ITS (Internal Transcribed Spacer, ribosomal region) marker.
The analysis conducted by use of thes markers showed that the coding regions rbcL and rpoC disply a low level of polymorphism and divergence; this resulted in a very low capacity of discriminating the different species both with Clustering and in Similarity methods.
This implies that the two regions seems to be less important as potential usefull barcode markers.
The region trnH-psbA chloroplast variability is much greater than previouses and therefore it is very interesting and promising as a barcode region with values of success in discriminating species, exceeding 75%.
The nuclear ITS region shows polymorphism and divergence levels much higher than all other markers, however, this elevated polymorphism and variability were not able to discriminate better than the space chloroplast region (trnH-psbA).
It is possible to assert that the results obtained are the same for both markers.
In conclusion, this first approach to the barcoding species which belong to the Italian wild maples showed encouraging results and highlighted speed, repeatibility and facility for a routine application of this method.
In force of the results here presented on Acer, we advise a possible application of this method to all other species of wild Italian dendroflora.
