Abstract
The abundance and diversity of Pythiaceous fungi assemblage on beech forest soils distributed along altitudinal and latitudinal gradients in Italy has been evaluated thought a traditional approach, based on the baiting technique, and massive sequencing analyses, by pyrosequencing (New Generation Sequencing-NGS). Both techniques revealed the presence of Pythiaceae in all the study sites. A total of 9 species (Pythium intermedium, Pythium macrosporum, Phytophthora cambivora, Phytophthora cactorum, Phytophthora plurivora and 4 additional undescribed Pythium) were detected with baiting, while sequences, resulted from the NGS analysis, analyzed with a custom database discriminated 26 Pythiaceae included species common to beech forest soils. The diversity and abundance indices of few species were found to be related to the altitude and the latitude of the sites. Pythium intermedium resulted associated to colder climate in northern Italy (Veneto latitudinal gradient) and high elevations (Latium altitudinal gradient) with the baiting technique. Similarly, according to 454 sequencing results, the presence of Pythium intermedium responded positively to latitude. In conclusion, the combination of these two methods resulted to be an efficient tool to determine a good bioindicator of climate change, such as Pythium intermedium.
