Abstract
The attention for environmental safety and the wide market requirement of wines able to express territory characteristics, increased the interest of researchers and viticulturist for local grape varieties. Vitis vinifera local germplasm can includes minor varieties which characteristics are often unknown with respect to genetic profile, viticultural and enological potential and level of resistance to biotic stresses, particularly respect the widespread fungal pathogens downy mildew (Plasmopara viticola) and powdery mildew (Erysiphe necator). The aim of this study was to characterize the susceptibility to downy and powdery mildew of five grape varieties belonging to the enological tradition of central Italy. Morphological features of leaf were analysed in the different varieties as possible natural defence mechanisms. Field observations and laboratory experiments on leaf disks and detached leaves were carried out to analyse the different susceptibility to the diseases. Epifluorescence microscopy was applied to study pathogens growth on leaf tissues and the content of stilbenes was assessed by HPLC in P. viticola inoculated leaves. Results showed significant differences among grape varieties with respect to susceptibility to P. viticola and E. necator. Trebbiano giallo and Romanesco varieties showed lower susceptibility to the pathogens compared to Aleatico and Canaiolo nero. Epifluorescence technique allowed us to determine differences among varieties with respect to the primary developmental stages of pathogen infections.
