Abstract: 
Si definisce cancerogena qualsiasi sostanza chimica in grado di alterare il DNA cellulare causando, spesso, danni irreversibili o mutazioni che possono portare all’insorgenza del cancro. L’uomo è continuamente esposto a tali sostanze che, presenti nell’ambiente, ritroviamo anche nel cibo e nell’acqua. Tra loro ricordiamo gli idrocarburi policiclici aromatici (IPA), una classe di composti altamente liposolubili che possono essere presenti sia in forma gassosa sia come particolato e sono caratterizzati, strutturalmente, dalla presenza di due o più anelli aromatici (anelli esagonali del benzene) condensati fra loro. Per poter interagire col DNA, questi composti, debbono essere attivati metabolicamente dando, così, origine a addotti voluminosi la cui presenza porta alla formazione d’accoppiamenti errati che, alla replicazione successiva, si convertiranno in mutazioni del tipo trasversioni e/o transizioni.
Per garantire l’integrità genomica, nel corso dell’evoluzione, le cellule hanno elaborato complessi sistemi enzimatici di riparazione in grado di riconoscere e rimuovere differenti tipi di lesioni. Queste vie metaboliche sono dette “Base Excision Repair” (BER), “Nucleotid Excision Repair” (NER), “Transcription Coupled Repair” (TCR), “Homologous Recombination Repair” (HRR) e “Non Homologous End-Joining” (NHEJ).
Lo scopo degli esperimenti da me condotti, in questi tre anni di dottorato, è stato quello di studiare quali di tali meccanismi di riparazione sono implicati nella rimozione delle lesioni indotte dagli idrocarburi policiclici aromatici ed, in particolare, quelli derivanti dalle forme metabolicamente attive (dioli epossidi) del benzo[a]pirene (BPDE) e del dibenzo[a,l]pirene (DBPDE). A tal proposito, la ricerca è stata condotta impiegando diverse linee cellulari di ovario di hamster Cinese tutte derivanti da una linea parentale comune (AA8) e ciascuna difettiva in uno dei meccanismi di riparazione sopra descritti (UV4, UV5, UV61, irs1SF, EM9 e V3-3).
Per questi esperimenti sono state utilizzate tecniche citogenetiche quali il test delle aberrazioni cromosomiche (AC) e quello degli scambi fra cromatidi fratelli (SCEs) Questi saggi sono internazionalmente riconosciuti validi per identificare la genotossicità di un composto. Abbiamo adottato due protocolli sperimentali, in uno dei quali, per l’analisi sia delle aberrazioni cromosomiche che degli SCEs, cellule asincrone sono state fatte crescere per circa un ciclo cellulare in presenza solo di BrdUrd, quindi trattate con differenti dosi della sostanza in esame e fissate a diversi tempi in base alla durata del loro ciclo cellulare, mentre nell’altro, che è il protocollo classico per l’analisi degli SCEs, cellule asincrone sono state trattate con differenti dosi della sostanza in esame e fatte crescere in continous per due cicli cellulari in presenza di BrdUrd e poi fissate a differenti tempi in base alla durata del loro ciclo cellulare.
L’ordine di sensibilità all’induzione delle aberrazioni cromosomiche, dopo trattamento con il BPDE, è risultato essere il seguente: UV4, irs1SF, UV5, UV61, EM9, V3-3 e AA8 in ordine decrescente, mentre per ciò che riguarda l’induzione degli SCEs l’ordine di sensibilità è risultato essere: UV5, UV4, V3-3, AA8, UV61, EM9 e irs1SF (in ordine decrescente), per ciò che riguarda gli SCEs indotti dal BPDE dopo un ciclo cellulare, e: AA8, UV5, UV4, UV61, V3-3, EM9 e irs1SF (in ordine decrescente), per ciò che riguarda gli SCEs indotti, dopo due cicli cellulari, in presenza del BPDE.
Dopo trattamento col DBPDE, invece, l’ordine di sensibilità risultante, nelle diverse linee cellulari, per ciò che riguarda le aberrazioni cromosomiche indotte da tale sostanza è stato: EM9, irs1SF, UV4, UV5, AA8, UV61 e V3-3, in ordine decrescente. Per gli SCEs indotti a seguito del trattamento col DBPDE per un solo ciclo cellulare, l’ordine di sensibilità è risultato essere il seguente: EM9, V3-3, UV61, UV4, AA8, UV5 e irs1SF (in ordine decrescente) e, dopo due cicli cellulari, invece, è risultato essere: V3-3, UV61, UV5, AA8, UV4, EM9 e irs1SF (in ordine decrescente).
Tali dati suggeriscono l’importanza del NER per la resistenza cellulare al BPDE nell’induzione sia d’aberrazioni cromosomiche sia degli SCEs. Inoltre, evidenziano un possibile ruolo della ricombinazione omologa nella resistenza cellulare al BPDE solamente per ciò che riguarda le aberrazioni cromosomiche e del NHEJ solamente per ciò che riguarda gli SCEs. Nel trattamento col DBPDE sembra, invece, che il BER sia essenziale per la resistenza cellulare a questo composto nell’induzione sia d’aberrazioni cromosomiche sia degli SCEs, mentre il NHEJ svolge un ruolo importante per ciò che riguarda la resistenza soltanto agli SCEs e l’HRR solamente alle aberrazioni cromosomiche.

Chemical agents that modify DNA causing irreversible damages or mutations, that can induce cancer, are said carcinogens. People are continually exposed to these substances that, being present in the environment, are also found in food and water . Amongst these agents we can find the polycyclic aromatic hydrocarbons (PAHs), a class of highly liposoluble compounds that can exist in both gaseous and solid form. They are characterized, structurally, by the presence of two or more aromatic rings condensates between themselves. To interact with DNA, these compounds must be metabolically activated originating bulky DNA adducts inducing formation of mismatch pairs that if not repaired, to the next replication, will cause mutations such as transversions and/or transitions. To ensure genomic stability, during evolution, cells elaborated several DNA repair mechanisms that detect and remove different types of lesions. These metabolic pathways can be summarize as follow: base excision repair (BER), nucleotide excision repair (NER), transcription coupled repair (TCR), homologous recombination repair (HHR) and non homologous end joining (NHEJ).
The aim of this work was to study which of these repair mechanisms is implicated in the removal of the lesions induced by polycyclic aromatic hydrocarbons (PAHs) and, specially, those arising by two metabolically activated intermediates (diol epoxides) of Benzo[a]Pyrene (BPDE) and Dibenzo[a,l]Pyrene (DBPDE).
Concerning this, the research was conducted employing different CHO cell lines (UV4, UV5, UV61,irs1SF, EM9 and V3-3) every deficient in one of the different DNA repair pathways above described and all arising from a common parental line, called AA8.
For this purpose we employed cytogenetic assays such as the study of induction of chromosomal aberrations (CA) and the sister chromatid exchanges (SCEs).
These two assays are well validated to detect genotoxic effects.
We found that the rank ordering of sensitivity for induction of chromosomal aberrations for BPDE, to be: UV4, irs1SF, UV5, UV61, EM9, V3-3 and AA8 (in descending order) and for induction of sister chromatid exchanges ,one cell cycle after the treatment was: UV5, UV4, V3-3, AA8, UV61, EM9 and irs1SF, whereas after second mitosis was: AA8, UV5, UV4, UV61, V3-3, EM9 and irs1SF (in descending order).
After DBPDE treatment, instead, the rank ordering of sensitivity for induction of chromosomal aberrations was: EM9, irs1SF, UV4, UV5, AA8, UV61 and V3-3 (in descending order)and for the induction of sister chromatid exchanges, one cycle after the treatment was found to be: EM9, V3-3, UV61, UV4, AA8, UV5 and irs1SF, whereas after two cell cycle was: V3-3, UV61, UV5, AA8, UV4, EM9 and irs1SF (in descending order).
These results suggest an important role of NER for cellular resistance to BPDE and of BER for cellular resistance to DBPDE, affecting both the inductions of chromosomal aberrations and sister chromatid exchanges. For both BPDE and DBPDE, HHR also seems to play a role in cellular resistance to these two compounds affecting mainly the induction of chromosomal aberrations, whereas NHEJ appears to play a role in cellular resistance to these two compounds, affecting only the induction of sister chromatid exchanges.
