Abstract: 
l lavoro svolto in questa tesi ha riguardato la caratterizzazione morfologico-ultrastrutturale
ed alla messa a punto di protocolli di crioconservazione degli spermatozoi di due specie di uccelli -
l’astore (Accipiter gentilis) ed il fagiano comune (Phasianus colchicus mongolicus). La prima,
l’astore, è un piccolo rapace selvatico, endemico in Europa, raramente mantenuto in cattività,
mentre la seconda, il fagiano, è una specie che viene allevata massivamente con successo per fini
commerciali già da diversi anni. Al momento le due specie non sono classificate fra quelle in
pericolo di estinzione. Tuttavia, l’astore lo potrebbe diventare data la rapidità con cui gli habitat
naturali e selvatici in Europa vengono modificati e antropizzati mentre il fagiano comune è il
rappresentante “domestico” del grande gruppo di fagiani in cui quelli cosiddetti “ornamentali” sono
a rischio di estinzione.
Queste sono le ragioni per le quali per queste specie vanno adottate delle misure di
salvaguardia. Due le principali proposte per la conservazione: in situ (protezione delle specie
all’interno del loro habitat), e ex-situ (allevamento in cattività). Nella seconda strategia un ruolo
fondamentale lo giocano la crioconservazione degli spermatozoi e l'inseminazione artificiale in
quanto permettono sia l'aumento degli individui sia il mantenimento della variabilità genetica di una
specie. Quando si vuole intraprendere il processo di crioconservazione il primo step di
fondamentale importanza è la caratterizzazione morfologica ed ultrastrutturale dei gameti maschili.
Infatti, in generale, specie animali diverse hanno spermatozoi con caratteristiche morfologiche ed
ultrastrutturali differenti, per ognuna delle quali devono attuarsi specifici protocolli di
crioconservazione. Quindi la prima parte di questo lavoro ha riguardato la caratterizzazione a livello
morfologico ed ultrastrutturale degli spermatozoi di queste due specie, mai descritti finora.
Attraverso le analisi alla microscopia elettronica è stato possibile osservare numerose analogie nella
morfologia e nell’ultrastruttura degli spermatozoi di astore e di fagiano e in base a quanto osservato
è stato possibile classificare i gameti maschili di queste due specie come appartenenti ai “non
passeriformi” poiché si sono chiaramente distinte la testa (con l’acrosoma e il nucleo) e la coda
(suddivisa in pezzo intermedio, principale e terminale). Altri elementi tipici degli spermatozoi “nonpasseriformi”
osservati negli spermatozoi di astore e di fagiano sono: la forma conica dell’acrosoma
e la presenza del perforatorium (contenuto nello spazio subacrosomiale che distalmente si inserisce
in un canale endonucleare nella regione anteriore del nucleo e che nel punto dove il nucleo si
innalza per formare il cosiddetto rostro nucleare presenta un ispessimento), dell’annulus (che
demarca il passaggio tra il pezzo intermedio e quello principale della coda), del centriolo prossimale
e di quello distale, del manicotto di mitocondri e delle nove fibre dense (localizzati a livello del
pezzo intermedio della coda), e della guaina fibrosa (posizionata lungo il pezzo principale della
coda). Le uniche differenze che sono state riscontrate negli spermatozoi di astore e fagiano sono la
lunghezza delle differenti regioni (testa e coda) e la lunghezza del centriolo distale. Quest’ultimo
infatti nel fagiano si estende per quasi l’intera lunghezza del pezzo intermedio mentre nell’astore
occupa solo una piccola parte di questa regione della coda. Sempre attraverso lo studio con la
microscopia elettronica sono state caratterizzate le categorie di anomalie morfologiche presenti nel
liquido seminale fresco, la cui analisi si è rivelata determinante per l’applicazione di tecniche di
conservazione degli spermatozoi. Infatti, è proprio la comparazione che permette di valutare gli
effetti della crioconservazione e fornisce indicazioni sulle eventuali modifiche da apportare ai
protocolli utilizzati. Dall’analisi quantitativa delle categorie spermatiche presenti negli eiaculati
freschi di entrambe le specie è emersa una percentuale di spermatozoi normali compresa in un
range che va dal 83,28% e il 90,28% nell’astore e dal 54,6% al 83,2% nel fagiano. La bassa
frequenza di spermatozoi anomali riscontrata è probabilmente fisiologica.
Una delle maggiori criticità quando si vuole intraprendere un processo di crioconservazione è la
disponibilità di campioni di eiaculati di buona qualità. Mentre nelle specie domestiche come il
fagiano questo problema è minimizzato dalla grande quantità di materiale a disposizione, in quelle
selvatiche è notevolmente amplificato dal limitato numero di individui che si riesce ad allevare in
cattività e dai ridotti volumi dei campioni di eiaculato raccolti. Questo lavoro è stato realizzato
grazie alla collaborazione con il centro di riproduzione assistita “European Falcons” di Assisi (Pg),
che ha partecipato alla sperimentazione fornendo i campioni di sperma fresco. Qui gli astori sono
allevati in cattività con il metodo cooperativo: i maschi imprintati sessualmente, se stimolati,
eiaculano volontariamente. Ottenere un esemplare di astore imprintato sessualmente richiede molto
tempo e lavoro per cui durante la sperimentazione è stato possibile utilizzare solo tre animali. I tre
maschi (denominati semplicemente A, B,C) erano tutti in età riproduttiva e all’inizio della
sperimentazione avevano 5 anni (maschio C), 6 anni (maschio B) e 7 anni (maschio A). Le
difficoltà scaturite dall’utilizzo di un numero limitato di animali sono state parzialmente attenuate
dai molteplici tentativi di campionamenti effettuati e dai numerosi campioni di eiaculati raccolti. I
tentativi di raccolta sono iniziati il primo marzo e sono terminati il 31 maggio di ogni anno e
nell’arco delle tre stagioni riproduttive con i tre animali presi in considerazione sono stati effettuati
in totale 216 tentativi di cui 178 sono andati a buon fine. Da questi ultimi sono stati ottenuti
campioni di spermatozoi non contaminati da feci e urine. I parametri utilizzati per valutare la qualità
degli campioni di eiaculati raccolti sono stati: volume, concentrazione, motilità, mobilità, vitalità,
percentuale delle anomalie morfologiche-ultrastrutturali, risultati ottenuti nel saggio di interazione
sperma-uovo e dati di fertilità nell’inseminazione artificiale. Dal confronto dei risultati ottenuti per
ogni parametro, l'individuo C è quello che ha mostrato i valori migliori. Il volume e la
concentrazione dei campioni di eiaculato raccolti, osservati nei tre maschi durante l’intero periodo
di sperimentazione, hanno mostrato i valori più alti nel mese di aprile, quindi a metà stagione
riproduttiva. La motilità e la vitalità degli spermatozoi sono risultate elevate durante l’intera
stagione riproduttiva; così come anche la mobilità, in particolar modo nel maschio B, che ha
mostrato una maggiore e più duratura capacità di movimento. Risultati altrettanto positivi si sono
ottenuti nel saggio di interazione sperma-uovo, che si basa sulla capacità degli spermatozoi di
penetrare la membrana perivitellina dell’uovo e fornisce indicazioni sulla loro capacità fecondante e
nell’inseminazione artificiale. Quindi questi risultati hanno dimostrato che con l’utilizzo del metodo
cooperativo è possibile raccogliere campioni di sperma utili per l’inseminazione artificiale e per i
processi di crioconservazione.
I fagiani di questo studio sono stati allevati presso il Dipartimento di Medicina Veterinaria,
Università di Pisa, ed erano alla loro prima stagione riproduttiva. I campioni di eiaculato sono stati
raccolti tramite massaggio dorso-addominale. Dal momento che questa specie viene allevata
massivamente già da diversi anni la qualità dei campioni di eiaculato freschi è stata presa in
considerazione solo esclusivamente come controllo positivo per valutare la bontà dei metodi di
crioconservazione applicati per questa specie e non per validare il metodo di raccolta come è stato
fatto per l’astore.
Per la crioconservazione dei gameti maschili di astore e fagiano sono stati impiegati tre diversi
protocolli di crioconservazione (denominati rispettivamente protocollo I, II e III). Nell’astore gli
eiaculati sono stati conservati singolarmente per ciascun esemplare (A,B,C) in modo da verificare
eventuali differenze nella capacità di risposta degli spermatozoi dei diversi animali al processo di
congelamento e scongelamento, mentre nel fagiano i campioni di spermatozoi raccolti sono stati
uniti in un pool e poi crioconservati. Complessivamente per l’astore sono stati utilizzati 46
campioni di eiaculato fresco, ciascuno dei quali è stato suddiviso in tre aliquote ad ognuna delle
quali è stato applicato uno dei tre protocolli per un totale di 138 prove di crioconservazione degli
spermatozoi di astore. Per il fagiano in dieci differenti giornate da dieci animali diversi sono stati
raccolti 100 campioni di eiaculato freschi. I campioni di ciascuna giornata sono stati raccolti in un
pool e crioconservati con il secondo protocollo (II). Il primo protocollo (I), che prevedeva l’utilizzo
del crioprotettore DMA (dimetilacetammide) in rapporto 1:1 rispetto al volume iniziale e del
prefreezing lake come diluente, è stato impiegato per la crioconservazione degli spermatozoi di
astore. Dopo l’ aggiunta del crioprotettore e del diluente, i valori di motilità e vitalità erano
estremamente bassi; al termine dell’intera procedura gli spermatozoi non sono risultati né motili né
vitali. All’analisi al SEM, gli spermatozoi appaiono completamente inviluppati in una fitta rete di
materiale, presumibilmente il DMA.
Il secondo protocollo (II) prevedeva l’utilizzo del DMA al 6% e del prefreezing lake come
diluente ed è stato utilizzato per la crioconservazione degli spermatozoi sia di astore sia di fagiano.
In questo caso, al termine del processo, nell’astore la percentuale degli spermatozoi vitali è risultata
in un range tra il 15% e il 69% e nel fagiano tra il 10,9% e il 48%. La percentuale di spermatozoi
con morfologia normale è risultata compresa in un range tra il 21,9% e il 29,9% nell’astore e tra il
6,1% e il 26,5% nel fagiano.
In ultimo, il terzo protocollo (III) prevedeva l’utilizzo del glicerolo come crioprotettore e del
prefreezing lake come diluente ed è stato impiegato per la crioconservazione degli spermatozoi di
astore. Già dopo la sola aggiunta del crioprotettore le percentuali di spermatozoi motili e vitali
erano molto basse, fino ad azzerarsi al termine della procedura. All’analisi al SEM gli spermatozoi
già nella fase di pre-congelamento hanno presentato molte anomalie, alcune delle quali irreversibili.
Quindi, sulla base dei risultati ottenuti si può affermare che, anche se sono necessarie ulteriori
modifiche, il protocollo che prevede l’utilizzo del prefreezing lake come diluente e del DMA al 6%
come crioprotettore può essere tenuto in considerazione per una eventuale applicazione nella
crioconservazione degli spermatozoi di astore e fagiano, che sebbene appartengano a ordini diversi,
hanno spermatozoi con una morfologia e un’ultrastruttura notevolmente simili. Pertanto
concludendo si può sostenere che due step di fondamentale importanza prima di intraprendere una
procedura di crioconservazione sono lo studio della morfologia e dell’ultrastruttura degli
spermatozoi e della qualità dei campioni di eiaculato freschi. L’analisi di quest’ultima oltre a fornire
delle indicazioni su quali campioni di spermatozoi sottoporre al processo di crioconservazione in
questo studio, per quanto riguarda l’astore, ha permesso anche di validare il metodo di raccolta.
I risultati ottenuti in questo lavoro, anche se preliminari, possono essere una buona base di partenza
per la messa a punto di metodologie di crioconservazione degli spermi anche in altre specie di
uccelli in modo da ampliare il campo di applicazione di questa metodica.

The present thesis focused on the development of protocols for sperm cryopreservation of two
species of birds - the goshawk (Accipiter gentilis) and the common pheasant (Phasianus colchicus
mongolicus). The goshawk is a diurnal raptor living in forest habitats, rarely bred in captivity. The
pheasant is a domestic species that has been successfully bred for commercial purposes for many
years. At the time of writing, these two species are not in danger of extinction. But the goshawk it
could become, given the speed with which the natural wild habitats in Europe are changing while
the common pheasant is representative of another genus of the Phasianidae family that is at risk of
extinction. These are the reasons why for these species should be adopted safeguard measures. In
order to protect endangered bird species two main proposals for conservation were formulated: in
situ, which provides for the protection of habitat to protect species at its internal, and ex-situ, which
relies on breeding and propagation in captivity. In this second strategy, the biotechnology of
reproduction, in particular the cryopreservation of semen and artificial insemination, plays a
fundamental role because it allows the increase in the number of individuals and the maintenance of
genetic variability. Before starting the cryopreservation process the ultrastructural analysis of the
male gamete is the first fundamental step; it also represents a database of reference in terms of
morphological categories present in normal semen to be taken into account when applying
techniques of sperm storage. In fact, in general, different animal species have spermatozoa with
different morphological and ultrastructural features, for each of which it must be accomplished
specific protocols of cryopreservation. So the first part of these work focused on the morphological
and ultrastructural sperm characterization of these two species, which has never been described so
far. The morphological features of the sperm cells evaluated in the present study allowed the
classification of the goshawk’s and pheasants’ spermatozoa as a non-passerine type, the more
“primitive” threadlike sperm type, characterized by five clearly defined segments: the acrosome and
the nucleus forming the head, and the midpiece, the principal piece and the endpiece constituting
the tail. Other typical elements of "non-passerines" spermatozoa observed in the goshawk and
pheasant sperm were: the conical acrosome and the presence of perforatorium (contained in a
subacrosomal space and distally inserted within a cylindrical endonuclear canal into the anterior
region of the nucleus and that showed a thickening at the point where the nucleus was raised to
form the so-called nuclear rostrum), of the annulus (which demarcated the boundary between the
midpiece and the principal piece), of the centriolar complex, of the mitochondrial sheath and the
nine dense fibers (located at the midpiece), and of the fibrous sheath (located along the principal
piece of the tail). The only differences found in the goshawk and pheasant sperm are the length of
the different regions (head and tail) and the length of the distal centriole. This latter in pheasant
stretches almost the entire length of the midpiece while in goshawk occupies only a small part of
this region of the tail. A description of the abnormal sperm categories in the goshawk’s and
pheasant’s fresh semen was also presented and the result of a quantitative analysis of normal and
abnormal spermatozoa indicated that a high percentage (ranged from 83,29% to 90,28% in goshawk
and from 56,4% and 83,29% in pheasant) of normal spermatozoa in fresh semen was indeed
present. The low percentage of abnormal sperm in the goshawks’ and pheasants’ fresh semen is
physiological. Upon undertaking a process of sperm cryopreservation, the key for success is to have
high quality samples of semen. While in domestic species such as pheasant this problem is
minimized by the large amount of material available in the wild species it is greatly amplified by
the limited number of individuals which can breed in captivity and by the reduced volumes of
samples of ejaculate collected. This research has been carried out with the collaboration of the
breeding center "European Falcons", Assisi, Italy. Their activities are based on innovative
principles and they collaborated for the entire duration of the experimentation, providing samples of
fresh semen which have then been used to perform various analysis. In this centre, goshawks are
bred in captivity using the cooperative method which offers many advantages, as it involves the use
of imprinted males which are much quieter than non-imprinted ones. In this particular type of
breeding centre, the males, isolated from conspecifics, socialize with and express sexual behaviours
towards the humans, spontaneously releasing the sperm when conveniently stimulated by them. To
get a goshawk imprinted sexually it requires much time and effort and so during the
experimentation it was possible to use only three animals. Although this limited number of
goshawks, the numerous collections (178) during three reproductive seasons allowed to analyze a
significant number of specimens. Males were evaluated for three consecutive reproductive seasons.
They were born in 2005 (male A), 2006 (male B) and 2007 (male C), thus were 4, 5 and 6 years old
at the beginning of the experimental period respectively. The characteristics of the goshawk’s
semen were assessed in order to optimize the breeding techniques for this species and to verify the
quality of the sperm obtained by using this method. In order to reach this goal, semen volume,
spermatozoa concentration, sperm cell motility, viability, sperm morphology, results in sperm-egg
interaction assay and data concerning artificial insemination were studied using semen collected
from three animals. The obtained results indicated that the youngest goshawk (male C) had the best
semen characteristics. As for semen volume and spermatozoa concentration, both observed in the
three males studied during the entire experimental period, the highest values were identified in the
middle reproductive season. Motility and viability of sperm were high throughout the breeding
season; as well as also the mobility. Regarding mobility, the sperm cells of the male B showed
lower values when compared to male C’s; however, their capacity to move forward, not only month
after month, but also as the time inside the Accudenz solution increased, resulted better in male B,
lasting for a longer period as well. Equally positive results were obtained in the sperm-egg
interaction assay, which is based on the ability of the sperm to penetrate the inner perivitelline layer
of eggs and provides guidance on their fertilizing capacity in artificial insemination. So these results
have shown that with the use of the cooperative method it is possible to collect sperm samples
useful for artificial insemination and for the processes of cryopreservation. The pheasants analysed
in this study were bred at the Department of Veterinary Science, University of Pisa, and were on
their first breeding season. Ejaculates were collected via torso-abdominal massage. As this species
is bred massively since several years the quality of samples of fresh ejaculate was studied only
exclusively as a positive control to evaluate the goodness of the cryopreservation methods applied
to this species and not to validate the method of collection as it was made for the goshawk. The last
part of this work covered the development of protocols for cryopreservation of goshawks’ and
pheasants’ male gametes. For cryopreservation of goshawk and pheasant male gametes they were
employed three different cryopreservation protocols (referred to as Protocol I, II and III). In
goshawk the ejaculates were cryopreserved individually for each specimen (A, B, C) in order to
check any difference in the response capacity of the sperm of different animals to the process of
freezing and thawing. In the pheasant samples of sperm collected were united in a pool and then
cryopreserved. Overall, for the goshawk they were used 46 samples of fresh ejaculate, each of
which was divided into three aliquots to which was applied to one of three protocols for a total of
138 tests of sperm cryopreservation of goshawk. For the pheasant in ten different days were
collected 100 samples of fresh ejaculate from ten different animals. Samples of each day were
collected in a pool and cryopreserved with the second protocol (II). The first protocol (I), that
included the use of pre-freezing lake as diluent and DMA (dimethylacetamide) in the ratio 1:1
compared to the initial volume, was used for goshawk sperm cryopreservation. After the addition of
cryoprotectant and diluent, the values of motility and vitality were extremely low; after the whole
process of cryopreservation, spermatozoa showed neither vitality nor motility. At SEM observations
spermatozoa appeared wrapped in the DMA that remained outside of the cellular elements. The
second protocol (II) involved the use of DMA at 6% compared to the initial volume and prefreezing
lake as a diluent and has been used for cryopreservation of spermatozoa both the goshawk and the
pheasant. After the whole cryopreservation process, the percentage of viable sperm anged from
15% to 69% in the goshawk and from 10.9% to 48% in the pheasant. The percentage of live normal
cells ranged from 21.9% to 29.8% in the goshawk and from 6.1% to 26.5% in the pheasant. Finally,
the third Protocol (III) involved the use of glycerol as a cryoprotectant and prefreezing lake and was
used as a diluent for goshawk sperm cryopreservation. Even after the addition of the cryoprotectant
the percentages of motile and viable sperm and at the end of the procedure spermatozoa showed
neither vitality nor motility. At SEM observation, the spermatozoa showed severe damage to the
different regions of the cells. Therefore we can state that Protocol II - involving the use of prefreezing
lake as diluent and the 6% DMA - can be held in high regard for sperm cryopreservation of
both the goshawk and the pheasant: even if they belong to different orders, they share a remarkably
similar morphology and ultrastructure of spermatozoa. Therefore concluding it can be argued that
two fundamental step before undertaking a cryopreservation process are the study of sperm
morphology and ultrastructure and quality of samples of fresh ejaculate. The analysis of this latter
as well as provide an indication about which samples of sperm submit to the cryopreservation
process in this study, with regard to the goshawk, allowed to validate also the method of collection.
The overall results of this study provide foundation for future studies to further develop a suitable
protocol for semen cryopreservation for both the species analysed and other species at risk of
extinction.
