Abstract: 
Le allergie causate dal frumento suscitano notevole attenzione in quanto sembrano
aumentate negli ultimi anni. In particolare, viene comunemente ritenuto che la causa di ciò
sia stata l’introduzione delle varietà create dopo la Rivoluzione Verde (capaci di dara
un’elevata produttività con apposite pratiche agricole, tra cui la fertilizzazione) rispetto alle
varietà tradizionali, le quali furono costituite per dare una produzione costante anche se
non eccellente, in condizioni ambientali variabili. In questo contesto si è inserito il presente
lavoro di dottorato che ha avuto due obiettivi principali: i) Stabilire il ruolo della
concimazione azotata su alcuni degli aspetti legati alle proprietà tecnologiche degli impasti
derivati dalle farine di frumento di nuova e antica costituzione. ii) Verificare se i livelli di
concimazione azotata più alti contribuiscano all’accumulo di proteine allergeniche, in
particolare quelle correlate con le allergie respiratorie, in modo diverso tra le varietà di
antica e di nuova costituzione. Sono state utilizzate due varietà di frumento duro di vecchia
costituzione, Senatore Cappelli e Urrìa, e due di nuova costituzione, Svevo e Claudio,
sottoposte a un triplo regime di concimazione azotata, in un’unica località in due annate
consecutive. Rispetto al primo obiettivo, è stato confermato un effetto positivo
sull’accumulo delle proteine totali nella cariosside di frumento su tutte le varietà, sia di
antica che di nuova costituzione, ma un effetto opposto tra le varietà di nuova e antica
costituzione per quanto riguarda l’accumulo della frazione gluteninica, nonché dei
parametri correlati alla forza dell’impasto. Per quanto riguarda il secondo obiettivo, è stato
osservato che la quantità di polipeptidi immunogenici correlati con le allergie respiratorie,
diminuisce con l’incremento del trattamento azotato in tutte le varietà considerate, sia
relativamente alla frazione metabolica che per la frazione CM-like (quest’ultima
maggiormente correlata con tale patologia). Inoltre, è stata confermata anche la già nota
alta variabilità della risposta soggettiva. L’analisi proteomica infine ha mostrato che la
concimazione azotata determina un maggiore nuemero di polipeptidi appartenenti alle
frazioni metaboliche e CM-like al regime superiore di fertilizzazione azotata e che le
proteine differenzialmente espresse corrispondono, nella maggior parte dei casi, ad
allergeni noti. Quindi, questi risultati mostrano che, almeno nei genotipi qui considerati e
nelle condizione prese in esame, non ci sono dati a favore dell’ipotesi che i genotipi di
vecchia costituzione contengano quantità inferiori di allergeni, sebbene il ruolo della
concimazione azotata nel miglioramento dei parametri qualitativi delle varietà di nuova
costituzione sia stata confermata.

Wheat allergy seems increased in recent years, and it is generally believed that this
is mostly due to the introduction of the new wheat cultivars developed after the Green
Revolution, that give a high yield with appropriate agricultural practices, included
fertilization, compared to traditional cultivars, that instead give constant production in a
variable environment, although not excellent. This work had two main objectives: i) to
establish the role of nitrogen fertilization on some of the aspects related to the
technological performance of wheat doughs based on new and old generation durum wheat
cultivars. ii) to verify if there are differences between old and new durum wheat varieties
in regard to the accumulation of allergenic proteins, in particular those related to
respiratory allergies, in relation to the amount of nitrogen used as fertilizer. Two traditional
(old generation) durum wheat cultivars, Senatore Cappelli and Urrìa, and two new
generation cultivars, Svevo and Claudio, submitted to three regimens of nitrogen
fertilization in a single location in two consecutive years, have been used. As for the first
objective, it was confirmed a positive effect on the accumulation of total proteins in wheat
kernel of all cultivars, but a divergent effect was observed between new and old generation
wheats with regard to the accumulation of the glutenin fraction, as well to parameters
related to the dough strength. With regard to the second objective, it has been observed that
the amount of immunogenic polypeptides involved in respiratory allergies, decreases with
the increase of the nitrogenous treatment in all varieties considered, both for the metabolic
and the CM-like fractions, this latter mostly correlated with this pathology. Furthermore, it
was also confirmed the already known high variability of the subjective response. Finally,
proteomic analysis showed that the nitrogen fertilization determines the accumulation of a
greater number of polypeptides belonging to the metabolic and CM-like proteins when
using higher nitrogen fertilization, and that the differentially expressed proteins
correspond, in most cases, to known allergens. Thus, these results show that, at least in the
genotypes here considered and in the conditions taken into consideration, there is no data
in favor of the hypothesis that the genotypes of the old generation contain lower amounts
of allergens, but the role of nitrogen fertilization in improving qualitative parameters of the
newly developed cultivars has been confirmed.
