Title: “Gene pyramiding” via ingegneria cromosomica di geni di resistenza a fusariosi e ruggini trasferiti in frumento tenero e duro da specie selvatiche del genere Thinopyrum

Abstract: 
Favorite dai cambiamenti climatici, le ruggini e la fusariosi della spiga (FHB), gravi malattie del frumento in tutto il mondo, sono diventate una minaccia anche in aree di distribuzione insolite. Pertanto, si rende necessaria l’introduzione di validi geni di resistenza in varietà adattate alla coltivazione, in modo da mantenere la loro capacità di resa e la qualità della granella. Il trasferimento di cromosomi estranei o di parte di essi nel genoma di frumento, definito “ingegneria cromosomica”, è una strategia di miglioramento che, combinata con l’utilizzazione di sistemi avanzati di selezione, permette di accedere ed impiegare nel breeding geni derivanti da specie selvatiche affini al frumento. Attraverso tale strategia, un gene/QTL di resistenza all’FHB (FHBres7el2), presente nell’accessione della Triticina selvatica Thinopyrum ponticum contenente cromatina el2, è stato trasferito da una linea di frumento tenero (KS24, 7DS.7el2L) in genotipi di frumento tenero (T4, 7DS.7DL/7el1L) e duro (7AS.7AL/7el1L), già portatori di altri geni utili, Lr19+Sr25+Yp, precedentemente trasferiti da una diversa accessione (el1) dello stesso Th. ponticum. Allo stesso modo, anche un altro gene/QTL di resistenza all’FHB (FHBres7E), presente in un’ altra specie dello stesso genere, il Thinopyrum elongatum, è stato oggetto di un altro trasferimento da una linea di frumento tenero, CS7E(7D), in genotipi di frumento tenero (T4, 7DS.7DL/7el1L) sempre in associazione ai geni Lr19+Sr25+Yp. Le progenie di incrocio derivanti da tali linee sono state caratterizzate e selezionate tramite l’ausilio di marcatori molecolari e tecniche di ibridazione in situ (GISH). Sebbene con diversa frequenza, sia in frumento tenero che in duro è stato possibile ottenere il pyramiding dei vari geni delle due accessioni di Th. ponticum (el1 + el2); nei frumenti teneri sono stati associati i geni provenienti dalle due specie Th. ponticum e Th. elongatum (el1 + E). Inoltre, sono stati sottoposti ad infezione con Fusarium spp., genotipi ricombinanti di tenero (7DS.7DL-7el17el2L) ed di duro (7AS.7AL-7el17el2L) per valutare l’efficacia dell’FHBres in entrambe le specie. Le linee selezionate, portatrici della nuova combinazione el1 (Lr19+Sr25+Yp) + el2 (FHBres), hanno in tutti i casi evidenziato una significativa riduzione (60-70%) della suscettibilità alla fusariosi della spiga.

Favoured by climatic changes, Fusarium Head Blight (FHB), a serious worldwide wheat disease, has become also a threat in unusual distribution areas. Therefore, new resistant and adapted varieties are needed, capable of maintaining their yield capacity and grain quality. The
transfer of alien chromosomes or fragments of them in the genome of wheat, defined as “chromosomal engineering”, is an improvement strategy which, combined with the use of advanced selection systems, allows to access and exploit genes derived from wild wheat relatives in breeding programs. Through this strategy, a gene/QTL for FHB resistance (FHBres7el2), present in the wild Thinopyrum ponticum accession that contains el2 chromatin, has been transferred from a bread wheat line (7DS.7el2L) to bread (7DS.7DL/7el1L) and durum wheat (7AS.7AL/7el1L) genotypes, already carrying other useful genes, i.e. Lr19+Sr25+Yp, previously transferred from a different (el1) Th. ponticum accession. Similarly, even another gene/QTL of FHB resistance (FHBres7E), present in a species of the same genus, the Thinopyrum elongatum, has been object of another transfer from a bread wheat line, CS7E(7D), to bread wheat (7DS.7DL/7el1L) genotypes, always in association with genes Lr19 + Yp + Sr25. In this thesis, with the help of molecular markers and in situ hybridization (GISH) techniques, the cross progeny of such lines were characterized and selected. Although with a different frequency, pyramiding of the Th. ponticum genes (el1 + el2) has been achieved in both bread and durum wheat; while the pyramiding of the Th. ponticum and Th. elongatum genes (el1 + E) has been achieved in the bread wheat. Also, to assess the FHBres efficacy in both species most of the bread (7DS.7DL-7el17el2L) and durum (7AS.7AL-7el17el2L) wheat recombinants were subjected to infection with Fusarium spp. The selected lines, carrying the novel el1 (Lr19+Sr25+Yp) + el2 (FHBres) combination, showed in all cases a significant reduction (60-70%) of the susceptibility to Fusarium Head Blight.
