Abstract: 
La presenza di temperature ambientali elevate determina una condizione di stress nella
bovina da latte con effetti negativi sulla salute, sulla produttività e sull'efficienza riproduttiva.
La prima attività ha riguardato la genotipizzazione di 446 bovine di razza Holstein per quattro
polimorfismi del singolo nucleotide (SNPs) dell’Hsp70.1: g895 C/- e g1128 G/T nella regione 5’-
UTR. Le PBMC sono state isolate da campioni di sangue, poste a 43°C in bagno termico per 1h e
poi incubate a 39°C in un’atmosfera contenente il 5% di CO2 per 1, 2, 4, 8, 16 e 24 ore. La vitalità
delle cellule è stata determinata utilizzando il saggio XTT. L’espressione del gene e della proteina
Hsp70.1 è stata determinata tramite real-time PCR e test ELISA, rispettivamente. I genotipi -- e TG
hanno mostrato la più alta (P<0.05) vitalità rispetto ai CC e GG, rispettivamente. I genotipi C- e TT
hanno fatto registrare una vitalità intermedia. L’espressione genetica dell’Hsp70.1 ha mostrato i
livelli più alti (P<0.001) nei genotipi -- e TG rispetto alla loro controparte. I genotipi -- e TG hanno
mostrato i livelli più alti della proteina inducibile Hsp70.1 rispetto a C-, TT e CC, GG. Questi
risultati indicano che la presenza di SNPs (C/- e G/T) nella regione 5’-UTR dell’Hsp70.1 migliora
la risposta allo shock termico e la tolleranza al caldo delle PBMC bovina. La seconda attività è stata
rivolta all’allestimento di un sistema di previsione (allerta) del rischio di stress da alte temperature,
specifico per la bovina da latte. Implementando la formula del calcolo dell’indice bioclimatico
Temperature-Humidity Index (THI) in funzione delle previsioni di temperatura e umidità relativa, è
stato possibile ottenere delle mappe di previsione dell’indice di stress termoigrometrico.

High environmental temperature, affect health, productivity and reproductive efficiency of
dairy cows.
446 Italian Holstein cows were genotyped for two Hsp70.1 single nucleotide polymorphisms
(SNPs): g895 C/- and g1128 G/T in 5’-UTR. 41 cows were selected to represent the different
genotypes. PBMC were isolated from blood samples and heated at 43°C in thermal bath for 1h and
then incubated at 39°C in atmosphere of 5% CO2 for 1, 2, 4, 8, 16 and 24h. Cell viability, gene and
protein expression of Hsp70.1 were determined. Genotypes -- and TG showed higher (P < 0.05)
viability compared with CC and GG, respectively. Genotypes C- and TT had intermediate viability.
Gene expression of Hsp70.1 showed higher (P < 0.001) levels in -- and TG genotype compared with
their counterparts. Genotypes -- and TG showed the higher level of inducible Hsp70.1 protein in
respect to C-, TT and CC, GG. These results indicate that the presence of SNPs (C/- and G/T) in the
5’-UTR region of inducible Hsp-70.1 ameliorates HS response and tolerance to heat of bovine
PBMC.
In the last years, Italian climatic conditions were characterized by an increase of intensity
and frequency of extreme events. To evaluate the heat discomfort on livestock, this study developed
an innovative Italian Forecasting Heat Warning System, using the THI-Temperature Humidity
Index, that is commonly used to quantify the degree of heat stress in farm animals. The system
provides six days forecast Italian maps of THI (day and night time) with indications of four hazard
levels to alert negative effects on production, health and welfare in dairy cows. This system
represents a valuable support to help farm managers.
