Abstract: 
Le scienze “omiche” genomica, proteomica e metabolomica sono
approcci complementari utilizzati per dare una completa visione
dell’intero sistema biologico. In questo lavoro di tesi è stato affrontato
l’approccio metabolomico, ovvero lo studio dei profili metabolici di
semi derivati dalla varietà di mais transgenico PR33P67 e della
corrispondente varietà tradizionale PR33P66 usando tecniche NMR
mono-bidimensionali, in modo tale da fornire informazioni sullo stato
attuale del sistema definendo il reale fenotipo.
La tecnica d’elezione, scelta per l’indagine metabolomica è la
Risonanza Magnetica Nucleare, poichè ci permette di ottenere dati
qualitativi e quantitativi di molti metaboliti che costituiscono il
complesso biologico; inoltre risulta essere una tecnica non distruttiva
che consente, contemporaneamente, la rilevazione dei dati in tempi
rapidi.
Studiare il profilo metabolico dei semi GM PR33P67 è di particolare
interesse, poiché i prodotti del mais sono un’importante fonte per i
nutrienti ed anche una parte essenziale della dieta umana e dei cibi
animali (entrambi usati attraverso diretto consumo).
La varietà transgenica contiene un gene funzionale Cry1A(b), che
conferisce alla pianta l’abilità di produrre la tossina insetticida Bt.
Negli estratti del seme di mais sono stati identificati circa quaranta
metaboliti idrosolubili per dare una più completa assegnazione dell’1H e
del 13C rispetto all’assegnazione riportata in letteratura. Per la prima
volta sono stati identificati nello spettro 1H NMR di estratti di semi di
mais i seguenti metaboliti: etanolo, acido lattico, acido citrico, lisina,
arginina, glicin-betaina, raffinosio, trealosio, -galattosio e l’adenina.
Gli spettri protonici di campioni di semi di mais transgenici e non2
transgenici risultano essere conservativi, poiché mostrano gli stessi
segnali e di conseguenza gli stessi metaboliti. Tuttavia, è stato osservato
che negli estratti transgenici rispetto ai non-transgenici vi è una più
elevata concentrazione di etanolo, acido citrico, glicin-betaina, trealosio
cosiccome in altri composti non ancora identificati. Di conseguenza, è
stato possibile discriminare i profili metabolici tra la varietà transgenica
e non-transgenica attraverso l’uso di una appropriata analisi statistica.
L’approccio metabolomico (come l’approccio genomico e proteomico)
ha avuto molto successo nell’ambito scientifico, diffondendosi anche
nel settore alimentare, soprattutto in cibi transgenici, poiché è necessario
determinare la sicurezza e la salubrità del prodotto.

The “omic” sciences, genomics, proteomics, and metabolomics, are
complementary approaches which are used in order to give a complete
vision of the entire biological system. In this thesis the metabolomic
approach, that is the deepening of the metabolic profiles of seeds
derived from the transgenic maize variety PR33P67 and the
corresponding traditional variety PR33P66, has been used. One- and
two-dimensional NMR techniques were used, in order to give
information about the actual status of the system by defining the real
phenotype. The Nuclear Magnetic Resonance is the technique of choice
for the metabolic investigation, because it allows us to obtain
identification and quantification data of many metabolites which
determine the biological complex; besides it is a technique that is not
destructive but it permits, at the same time, to detect the data in short
time.
The GM maize seed metabolic profile is of particular interest because
maize products are an important source of nutrients and, therefore, an
essential part of the human diet (both through direct consumption and
animal feeds). The transgenic variety carries a functional Cry1A(b)
gene, which confers to the plant the ability to produce Bt insect toxin.
About forty water-soluble metabolites in the maize seed extracts were
identified, providing a more complete 1H and 13C NMR assignment with
respect to the assignment reported in the literature. In particular,
ethanol, lactic acid, citric acid, lysine, arginine, glycine-betaine,
raffinose, trehalose, -galactose, and adenine were identified for the first
time in the 1H NMR spectrum of maize seed extracts. The 1H spectra of
transgenic and non-transgenic maize seed samples turned out to be
conservative, showing the same signals and therefore the same
metabolites. However, a higher concentration of ethanol, citric acid,

glycine-betaine, trehalose, as well as of another compound not yet
completely identified, was observed in the transgenic extracts than in
non-transgenic samples. So, it was possible to discriminate between
transgenic and non-transgenic metabolic profilings through the use of an
appropriate statistical analysis.
The metabolic approach (as well as the genomic and proteomic
approach) has had a great success in the scientific research and also in
the nutrition field, above all in transgenic food, because it is important
to determine the health status and safety of the product.
