Title: Indagini epidemiologiche, fenotipiche e genotipiche di popolazioni del fungo Cryphonectria parasitica nel comprensorio dei Monti Cimini

Abstract: 
Cryphonectria parasitica, agente casuale del cancro corticale del castagno, è da anni causa
principale del declino dei castagneti nel comprensorio dei Monti Cimini. Dagli studi condotti negli
ultimi 20 anni si è notata una stabilizzazione del fattore ipovirulenza sul territorio, con un notevole
rallentamento dello stato epidemico della malattia. Tuttavia, lo studio della compatibilità vegetativa,
ha evidenziato la presenza di sei gruppi di compatibilità presenti tipicamente, alcuni solo nel nord
Italia, ed altri i solo nel Sud Italia e recanti ciascuno cinque loci polimorfici, caratteristica che, in
caso di riproduzione sessuata potrebbe portare alla comparsa di nuovi gruppi di compatibilità con
una probabile recrudescenza della malattia.
Gli scopi del presente lavoro sono: valutare la capacità riproduttiva del fungo in campo,
attraverso la verifica della produzione di strutture di riproduzione gamica e della loro vitalità;
determinare il numero dei gruppi di compatibilità presenti nella progenie dei periteci e studiare la
variabilità genetica all’interno di questi; è stata infine analizzata la presenza di entrambi i tipi
sessuali Mat-1-1 e Mat1-2 evidenziando l’eventuale presenza di heterocaryon.
I risultati ottenuti indicano una costante presenza di stroma con periteci su cancri “letali”.
L’analisi della vitalità dei periteci, svolta in ambiente controllato, ha evidenziato una spiccata
capacità di maturazione dei periteci e di espulsione delle ascospore prodotte. La quantificazione
delle ascospore e la loro capacità di germinazione si sono però mostrate in tutti i casi limitate.
L’analisi delle prove di compatibilità tra la progenie dei periteci ottenuta, ha evidenziato
segregazione tra tutte le progenie testate. Dall’analisi dei test di compatibilità svolti, è emersa una
distribuzione analoga a quella ottenuta da lavori precedenti. Per quanto riguarda l’analisi dei tipi
sessuali si evidenzia la presenza di entrambi i tipi sessuali, Mat1-1 e Mat1-2 nella maggior parte dei
periteci analizzati, evidenziando inoltre la presenza di heterocaryon. I risultati ottenuti indicano una
situazione ancora stazionaria ma, la presenza di strutture di riproduzione sessuata e di entrambi i tipi
sessuali mantengono alto il rischio di ripresa della malattia, rischio che aumenterebbe
esponenzialmente in caso di introduzione di ulteriori gruppi di compatibilità esogeni.
Si auspica quindi un controllo delle attività collegate all’industria boschiva e una
regolamentazione della circolazione di legname tra aree con popolazioni del fungo differenti.

Cryphonectria parasitica, the causal agent of the chestnut blight, is one of the major disease of
chestnut trees worldwide also the cause of the decline of the chestnut in the Monti Cimini areas.
The frequency of normal and altered phenotypes did not changed consistently in the last 20 years,
suggesting that sexual reproduction is strongly limited or absence. However, the study of vegetative
compatibility revealed the presence of six groups of compatibility present typically, some only in
Northern Italy and others in South Italy alone and bearing each five polymorfic loci feature that, in
the case of sexual reproduction potentially would allow the differentiation of new v-c groups in the
investigated populations. The aims of this work are: assessing the reproductive ability of the fungus
in field, through the verification of the production of gamic reproduction and their vitality structures
determining the number of present compatibility in the offspring of the perithecia groups and study
the genetic variability; the presence of both sexual mating type was finally analysed and the
presence of heterocaryon.
Results indicate a constant presence of stromata with perithecia on "deadly cancers. The analysis of
the vitality of the perithecia, showed a remarkable capacity of maturation of the peritechia and
expulsion of the produced ascospores. The quantification of the ascospores and their ability to
germination have however shown in all cases limited. The vegetative compatibility tests between
the offspring of the perithecia obtained, showed segregation between all the tested progeny and the
analysis of vegetative compatibility European groups showed a similar distribution to that obtained
from previous work. Both mating types were found in all perithecia, making potentially possible
recombination trought sexual reproduction. The majority of the analysed perithecia also shows the
presence of heterocaryon. Results indicate a still stationary situation, but the presence of sexual
reproduction and sexual both structures maintain high risk of resurgence of the disease to the
differentiation of new vc-types trought sexual recombination or introduction from new areas.
Hopefully then a regulation throught forestry activities and trading of chestnut products between
areas with different populations of the fungus.
