      <title>The intergenic spacer region of the rDNA of Olea europaea</title>
      <link>http://hdl.handle.net/2067/1308</link>
      <description>Title: The intergenic spacer region of the rDNA of Olea europaea
Authors: Maggini, Fabio; Gelati, Maria Teresa; Spolverini, Michela; Frediani, Manuela
Abstract: The intergenic spacer (IGS) region of rDNA in&#xD;
Olea europaea is 5,629 bp long and it can be subdivided,&#xD;
on the basis of the different features, into six regions; two&#xD;
of them, located upstream and downstream of the putative&#xD;
transcription initiation site (TIS), respectively, contain&#xD;
prominent and unrelated subrepeats (s.r.). The upstream s.r.&#xD;
are 75–86 bp in length and the downstream ones are about&#xD;
160 bp in length. The first type is represented by a high number&#xD;
(36) of iterations, which share a mutual similarity of 82.5%. A&#xD;
single sequence (TATTATAGGGGGGAGG) is found, which&#xD;
fits the reported TIS of plants and the position +1 would&#xD;
correspond to the adenine at position 3,622 after the 3′ end of&#xD;
25 S ribosomal RNA (rDNA) genes. Moreover, in Olea IGS&#xD;
region some unusual features are observed and discussed in&#xD;
relation to analogous data present in the literature.

      <title>Phylogenetic relationships among annual and perennial species of the genus Cicer as inferred from ITS sequences of nuclear ribosomal DNA</title>
      <link>http://hdl.handle.net/2067/1309</link>
      <description>Title: Phylogenetic relationships among annual and perennial species of the genus Cicer as inferred from ITS sequences of nuclear ribosomal DNA
Authors: Frediani, Manuela; Caputo, Paolo
Abstract: The cladistic analysis of the DNA sequences of the internal transcribed spacers of ribosomal cistrons (ITS1 and ITS2) for 20 species of Cicer L. (among which all the annuals), shows that various sections of the genus are not monophyletic. Annual species do not form a clade: C. arietinum, in fact, is closely related to both C. echinospermum and C. reticulatum, whereas C. bijugum, C. judaicum, and C. pinnatifidum form a separate clade. The annual C. cuneatum is sister group to the perennial C. canariense and both are archaic species within the genus. C. yamashitae is, on the contrary, the only annual species belonging to a group of perennials, within which close relationships are evident between C. graecum and C. montbretii as well as among a group of mainly Asian species.

      <title>Cytology of Vicia species. X. Karyotype evolution and phylogenetic implication in Vicia species of the sections Atossa, Microcarinae, Wiggersia and Vicia</title>

      <description>Title: Cytology of Vicia species. X. Karyotype evolution and phylogenetic implication in Vicia species of the sections Atossa, Microcarinae, Wiggersia and Vicia
Authors: Ruffini Castiglione, Monica; Frediani, Manuela; Gelati, Maria Teresa; Ravalli, Concetta; Venora, Gianfranco; Caputo, Paolo; Cremonini, Roberto
Abstract: Automated karyotype analyses and sequence of&#xD;
rDNA spacers have been analysed for the species belonging&#xD;
to sections Atossa, Microcarinae, Wiggersia and Vicia.&#xD;
Karyomorphological parameters, based on Rec, Syi and TF&#xD;
% indices, have been determined and evidenced that, in&#xD;
term of symmetry, the karyotype of Vicia lathyroides was&#xD;
the most asymmetric one. A multivariate analysis using 34&#xD;
karyological parameters, in addition to the symmetry&#xD;
indices, has been carried out and the corresponding&#xD;
dendrogram of linkage distances showed six different&#xD;
groups. Molecular investigations on the inclusive group in&#xD;
study by employing ITS DNA sequences indicated a&#xD;
different pattern of relationships. The cladistic analysis&#xD;
combining the molecular data set with karyological parameters&#xD;
evidenced that the species of sections Vicia and Atossa&#xD;
join closely to each other in a paraphyletic group, which&#xD;
includes the monophyletic section Wiggersia. Therefore,&#xD;
our karyological and molecular data provide information&#xD;
about the phylogenetic position of the analysed species&#xD;
inside the subgenus Vicia and are discussed in relation to&#xD;
previous results obtained by morphology, isozymes and&#xD;
ribosomal genes analyses.

      <title>Nuclear DNA contents, rDNAs and karyotype evolution in subgenus Vicia. III. The heterogeneous section Hypechusa</title>

      <description>Title: Nuclear DNA contents, rDNAs and karyotype evolution in subgenus Vicia. III. The heterogeneous section Hypechusa
Authors: Caputo, Paolo; Frediani, Manuela; Venora, Gianfranco; Ravalli, Concetta; Ambrosio, Maria; Cremonini, Roberto
Abstract: Nuclear DNA contents, automated karyotype analyses, and&#xD;
sequences of internal transcribed spacers from ribosomal genes have been&#xD;
determined in the species belonging to section Hypechusa of the subgenus&#xD;
Vicia. Karyomorphological results and phylogenetic data generated&#xD;
from the comparison of rDNA (genes coding for rRNA) sequences&#xD;
showed that sect. Hypechusa is not monophyletic; however, some monophyletic&#xD;
units are apparent (one including Vicia galeata, V. hyrcanica,&#xD;
V. noeana, and V. tigridis, another including V. assyriaca, V. hybrida,&#xD;
V. melanops, V. mollis, and V. sericocarpa), which partly correspond to&#xD;
morphology-based infrasectional groups. The relationships among these&#xD;
species and the species in sections Faba, Narbonensis, Bithynicae, and&#xD;
Peregrinae have been also investigated.

      <title>Ampelographic and DNA Characterization of Local Grapevine Accessions of the Tuscia Area (Latium, Centre of Italy)</title>

Authors: Muganu, Massimo; Dangl, Gerald; Aradhya, Malli; Frediani, Manuela; Scossa, Angela; Stover, Ed
Abstract: The presence of local vines in the Tuscia area (Latium region, Italy) was documented by historical sources. Ampelographic and molecular characterization (35 morphological descriptors and 16 microsatellite loci) was carried out on accessions belonging to the most frequently mentioned local vines. SSR markers have enabled us to distinguish cases of local synonymy and microsatellite profiles have permitted comparison with previously reported profiles indicating synonymy and divergence with vines in other collections. Ampelographic traits showed that some accessions of the same genotype differ in minor characteristics that can have a role in clonal evaluation.</description>
